comt(catechol-o-methyltransferase)基因位于人類(lèi)第22號(hào)染色體q11.21上,在不同的細(xì)胞中被翻譯成不同長(zhǎng)度的蛋白。在大腦的神經(jīng)細(xì)胞中,comt基因表達(dá)為膜結(jié)合的兒茶酚-o-甲基轉(zhuǎn)移酶蛋白;大腦中的兒茶酚-o-甲基轉(zhuǎn)移酶蛋白主要分布在前額葉皮質(zhì)區(qū),它可以保證神經(jīng)遞質(zhì)如多巴胺等在一個(gè)穩(wěn)定的水平上。在肝、腎及血液等器官中表達(dá)為可溶性的兒茶酚-o-甲基轉(zhuǎn)移酶蛋白,它可以催化甲基從s-腺苷甲硫氨酸轉(zhuǎn)移到兒茶酚胺上,此類(lèi)物質(zhì)包括神經(jīng)遞質(zhì)多巴胺、腎上腺素、去甲腎上腺素等。comt基因在代謝治療高血壓、哮喘、帕金森疾病的兒茶酚藥物方面起著重要的作用。由于利培酮與多巴胺、5-羥色胺等多種神經(jīng)遞質(zhì)受體均有相互作用,因此尋找利培酮代謝與comt基因多態(tài)性的關(guān)系是現(xiàn)有技術(shù)急需解決的難題。

對(duì)現(xiàn)有文獻(xiàn)的檢索,未發(fā)現(xiàn)有本發(fā)明的comt基因snprs737865為利培酮分子標(biāo)記物的相關(guān)報(bào)道。現(xiàn)有的利培酮分子標(biāo)記研究多基于歐美人群,不能在中國(guó)漢族人群中應(yīng)用。而且由于精神分裂病人利培酮單藥治療后符合標(biāo)準(zhǔn)的病人較少,樣本收集十分困難,現(xiàn)有研究由于樣本量小,不具有相應(yīng)的代表性。而本發(fā)明通過(guò)擴(kuò)大樣本量在中國(guó)漢族人群中進(jìn)行大規(guī)模發(fā)掘和驗(yàn)證,發(fā)現(xiàn)了comt基因snprs737865可以作為利培酮分子標(biāo)記物,具有明顯的經(jīng)濟(jì)效益。



技術(shù)實(shí)現(xiàn)要素:

針對(duì)現(xiàn)有技術(shù)中的缺陷,本發(fā)明的目的是提供一種基于rs737865的檢測(cè)利培酮用藥效果的試劑盒;具體地講是一種分離核酸、一種等位基因特異性的核酸引物和comt基因snprs737865單核苷酸多態(tài)性的檢測(cè)方法。

本發(fā)明的目的是通過(guò)以下技術(shù)方案實(shí)現(xiàn)的:

第一方面,本發(fā)明提供了一種分離的含單核苷酸多態(tài)性位點(diǎn)的核苷酸,所述核苷酸的序列如seqidno:1所示,所述單核苷酸多態(tài)性位點(diǎn)位于第858位,rs737865為a→g。

第二方面,本發(fā)明提供了一種如前述的核苷酸在制備檢測(cè)利培酮用藥效果的試劑盒中的用途。

優(yōu)選的,snprs737865為利培酮分子標(biāo)記物。

第三方面,本發(fā)明提供了一種分離的核苷酸,所述核苷酸的序列如seqidno:1所示第858位,該位點(diǎn)為a。

第四方面,本發(fā)明提供了一種檢測(cè)利培酮用藥效果的試劑盒,所述試劑盒包括:根據(jù)如seqidno:1所示序列的第858位的上下游設(shè)計(jì)的引物對(duì)。

作為優(yōu)選方案,所述引物對(duì)為野生型引物對(duì)或突變型引物對(duì),其中,所述野生型引物對(duì)的序列如seqidno:3、seqidno:4所示:

正向引物:5′-acaataaaaagcaacaggacacaaaaaacc-3′seqidno:3

反向引物:5′-tgctgtgatgaggggctccagatag-3′seqidno:4;

所述突變型引物對(duì)的序列如seqidno:5、seqidno:4所示:

第五方面,本發(fā)明提供了一種如前述的試劑盒的非診斷目的使用方法,其包括如下步驟:

s1,提取樣品dna;

s2,利用所述引物對(duì)進(jìn)行擴(kuò)增,得擴(kuò)增產(chǎn)物;

s3,檢測(cè)樣品中是否存在如seqidno:1所示的單核苷酸多態(tài)性位點(diǎn)rs737865。

作為優(yōu)選方案,所述步驟s3采用dna測(cè)序、雜交測(cè)序、酶促錯(cuò)配切割、異源雙鏈分析、點(diǎn)雜交、寡核苷酸陣列、mini-sequencing、分子信標(biāo)中的任意一種方法。

作為優(yōu)選方案,步驟s3包括如下步驟:采用abi7700熒光定量pcr儀,對(duì)所取樣品的位點(diǎn)rs737865進(jìn)行分型,檢測(cè)第858位否存在單核苷酸多態(tài)性。

作為優(yōu)選方案,所述引物對(duì)的長(zhǎng)度為15-50bp。

作為優(yōu)選方案,步驟s2中,所述擴(kuò)增的具體條件如下:反應(yīng)體系總體積為20μl,其中:

100μmol·l-1dntps;

50mmol·l-1kcl;

10mmol·l-1tris-hcl,ph為8.3;

1.5mmol·l-1mgcl2:

0.5utaqdna聚合酶;

20-50ng模板dna;

0.5μl10pmseqidno.3;

0.5μl10pmseqidno.4;

雙蒸水補(bǔ)足至20ul;

pcr反應(yīng)條件:95℃2mins;30×(94℃30s;58℃30s,72℃30s)72℃5mins。

第六方面,本發(fā)明提供了一種用于檢測(cè)如前述的核苷酸的snp位點(diǎn)的野生型引物對(duì),所述引物對(duì)的序列如seqidno:3、seqidno:4所示:

正向引物:5′-acaataaaaagcaacaggacacaaaaaacc-3′seqidno:3

反向引物:5′-tgctgtgatgaggggctccagatag-3′seqidno:4。

第七方面,本發(fā)明提供了一種用于檢測(cè)如前述的核苷酸的snp位點(diǎn)的突變型引物對(duì),所述引物對(duì)的序列如seqidno:5、seqidno:4所示:

正向引物:5′-aaaaagcaacaggacacaaaaagcc-3′seqidno:5

反向引物:5′-tgctgtgatgaggggctccagatag-3′seqidno:4。

本發(fā)明采用的樣品來(lái)自上海市精神衛(wèi)生中心的995個(gè)精神分裂癥患者,年齡18~60歲。全體參與者均由家屬在正式的知情同意書(shū)上同意簽字。

在上述的基礎(chǔ)上,本發(fā)明采用片段長(zhǎng)度差異等位基因特異性pcr(fldas-pcr)分型技術(shù)對(duì)comt基因snprs737865位點(diǎn)及其頻率分布情況進(jìn)行了檢測(cè)。已經(jīng)發(fā)現(xiàn)了comt基因snprs737865第858位a→g多態(tài)性。

與現(xiàn)有技術(shù)相比,本發(fā)明具有如下的有益效果:

本發(fā)明為研究我國(guó)人群中comt基因多態(tài)性與臨床用藥安全的關(guān)系奠定了基礎(chǔ),為臨床用藥個(gè)體化提供理論基礎(chǔ),同時(shí)也為基于藥物基因組學(xué)理念的新藥研發(fā)提供指導(dǎo)依據(jù)。

具體實(shí)施方式

下面結(jié)合具體實(shí)施例對(duì)本發(fā)明進(jìn)行詳細(xì)說(shuō)明。以下實(shí)施例將有助于本領(lǐng)域的技術(shù)人員進(jìn)一步理解本發(fā)明,但不以任何形式限制本發(fā)明。應(yīng)當(dāng)指出的是,對(duì)本領(lǐng)域的普通技術(shù)人員來(lái)說(shuō),在不脫離本發(fā)明構(gòu)思的前提下,還可以做出若干變形和改進(jìn)。這些都屬于本發(fā)明的保護(hù)范圍。

下列實(shí)施例中未注明具體條件的實(shí)驗(yàn)方法,通常照常規(guī)條件,例如sambrook等人的分子克?。簩?shí)驗(yàn)室手冊(cè)(newyork:coldspringharborlaboratorypress,1989)中所述的條件,或按照制造廠(chǎng)商所建議的條件。

本發(fā)明涉及comt基因第二號(hào)內(nèi)含子snprs737865的單核苷酸多態(tài)性的檢測(cè)方法,同時(shí)涉及相關(guān)的分離核酸以及引物。本發(fā)明通過(guò)關(guān)聯(lián)分析發(fā)現(xiàn)了comt基因第二號(hào)內(nèi)含子snprs737865能夠作為利培酮的分子標(biāo)記物,該comt基因第二號(hào)內(nèi)含子的序列如seqidno:1所示,其中第858位存在單核苷酸多態(tài)性,即snprs737865。本發(fā)明針對(duì)該snprs737865設(shè)計(jì)了相關(guān)的引物對(duì)。

本發(fā)明采用fldas—pcr探針技術(shù)對(duì)comt基因的第二號(hào)內(nèi)含子snp位點(diǎn)及其頻率分布情況進(jìn)行了檢測(cè),發(fā)現(xiàn)comt基因的第二號(hào)內(nèi)含子第858位a→g多態(tài)性。

具體而言,本發(fā)明在comt基因第二號(hào)內(nèi)含子上的snprs737865上下游設(shè)計(jì)fldas-pcr引物,然后用聚丙烯酰胺凝膠電泳對(duì)snp位點(diǎn)rs737865進(jìn)行分型,發(fā)現(xiàn)comt基因第二號(hào)內(nèi)含子snp(即第858位a→g多態(tài)性)。

本發(fā)明有大量的分析技術(shù)可用于檢測(cè)comt基因第二號(hào)內(nèi)含子中所述位點(diǎn)是否存在單核苷酸多態(tài)性;這些技術(shù)包括(但并不限于):dna測(cè)序、雜交測(cè)序;酶促錯(cuò)配切割、異源雙鏈分析、點(diǎn)雜交、寡核苷酸陣列(dna芯片)、mini-sequencing、分子信標(biāo)等,變性高效液相色譜法(dhplc)。本發(fā)明的檢測(cè)方法被用于評(píng)估個(gè)體comt基因第二號(hào)內(nèi)含子snprs737865多態(tài)性與氯氮平代謝的關(guān)系。用于本發(fā)明的測(cè)試樣品沒(méi)有特別限制,對(duì)于檢測(cè)snp而言,可以是從分離的血液、組織等樣品中抽提出的dna或mrna;對(duì)于comt基因第二號(hào)內(nèi)含子活性而言,可以任何含有comt基因第二號(hào)內(nèi)含子的樣品,如分離的血液等。

所述的“分離出的或純化的”是指在基因組dna水平上,利用引物對(duì)來(lái)擴(kuò)增每個(gè)多態(tài)性位點(diǎn)的基因序列和利用純化試劑盒對(duì)擴(kuò)增產(chǎn)物進(jìn)行提純。

以下是對(duì)本發(fā)明方案的具體實(shí)施過(guò)程:

本實(shí)施例采用的樣品(具體為血液樣品)來(lái)自上海市精神衛(wèi)生中心的995個(gè)精神分裂癥患者,年齡18~60歲;全體參與者均由家屬在正式的知情同意書(shū)上同意簽字。本實(shí)施例涉及的comt基因第二號(hào)內(nèi)含子的序列見(jiàn)seqidno:1,其所編碼的氨基酸序列列于seqidno:2;comt基因第二號(hào)內(nèi)含子的基因組序列可以從genbank中獲得。

引物設(shè)計(jì)信息:

采用primer5.0軟件,以genbank數(shù)據(jù)庫(kù)comt基因(id:1312)第二號(hào)內(nèi)含子以及內(nèi)含子與內(nèi)含子接合部位序列為模板設(shè)計(jì)一對(duì)引物,由invitrogen公司合成。

引物信息,擴(kuò)增comt基因第二號(hào)內(nèi)含子snp位點(diǎn)的dna序列引物信息:

pcr擴(kuò)增條件:(體系各項(xiàng)試劑由abi公司提供,也可以從公開(kāi)的渠道獲得)

反應(yīng)體系總體積為20μl,其中:

100μmol·l-1dntps;

50mmol·l-1kcl;

10mmol·l-1tris-hcl,ph為8.3;

1.5mmol·l-1mgcl2;

0.5utaqdna聚合酶;

20-50ng模板dna;

0.5μl10pmseqidno.3;

雙蒸水補(bǔ)足至20ul;

pcr反應(yīng)條件:95℃2mins;30×(94℃30s;58℃30s,72℃30s)72℃5mins。

上述pcr反應(yīng)完成后,擴(kuò)增產(chǎn)物經(jīng)非變性聚丙烯酰胺凝膠電泳后銀染顯帶,可以發(fā)現(xiàn)comt基因snprs737865野生型(基因型為a)擴(kuò)增產(chǎn)物比突變型(基因型為g)多了5bp,因此基因型為純合個(gè)體擴(kuò)增產(chǎn)物為單一條帶,雜合個(gè)體為雙條帶。比較發(fā)現(xiàn)comt基因snprs737865有多態(tài)性(即第858位a→g)。關(guān)聯(lián)分析發(fā)現(xiàn)comt基因snprs737865的g基因型與a基因型患者用利培酮治療前后有顯著的的個(gè)體差異,p值為0.007(見(jiàn)表1)。

本發(fā)明為研究我國(guó)人群中comt基因多態(tài)性與臨床用藥安全的關(guān)系奠定了基礎(chǔ),為臨床用藥個(gè)體化提供理論基礎(chǔ),同時(shí)也為基于藥物基因組學(xué)理念的新藥研發(fā)提供指導(dǎo)依據(jù)。
