本發(fā)明涉及用于疫苗組合物中的多肽佐劑。
佐劑是一種物質(zhì)或方法,它可以通過調(diào)節(jié)免疫細胞的活性來特異地增強對抗原的免疫反應(yīng)。佐劑的例子包括弗氏佐劑,胞壁酰二肽,脂質(zhì)體。佐劑也可以是具有激動作用或拮抗作用的由免疫細胞表達的抗體直至受體。佐劑與載體的區(qū)別在于載體通常被用來增強對抗原的免疫反應(yīng)。
載體是一種具有免疫原性的分子,當它與另一個分子相結(jié)合時可以增強后者的免疫反應(yīng)。有一些本身不具有免疫原性的抗原(即在它們直接存在時不具有免疫原性),但當它們與諸如匙孔血藍素(keyhole-limpethemocyanin)或破傷風類毒素的外源蛋白分子相連時則可具有產(chǎn)生抗體反應(yīng)的能力。這些抗原含有B細胞表位而沒有T細胞表位。這些軛合物(“載體”蛋白)的蛋白部分提供T細胞表位來刺激輔助性T細胞,從而刺激抗原特異性B細胞分化為漿細胞,并產(chǎn)生針對于該抗原的抗體。為免疫細胞受體的蛋白配基的佐劑可以同時具有佐劑和載體的性質(zhì),而后者依賴于可被免疫系統(tǒng)T細胞識別的“外源”多肽序列的組成。本發(fā)明中的新型佐劑具有以上所述的兩種性質(zhì)。
多蛋白抗原或多梯狀(polyladder)蛋白是由多種寄生性或自由生活的線蟲產(chǎn)生的。這些多蛋白通常包括許多直接串聯(lián)排列的多亞基,這些蛋白通常作為大前體蛋白合成,其可被蛋白酶剪切產(chǎn)生大小間隔約為15KD的、“梯狀”的小片段,反應(yīng)單個功能域的平均分子量(denominatormolecular mass)的增加。每一個這樣亞基的最后4個氨基酸都具有一種易被蛋白酶消化的RX(K/R)R(或有時是RXFR)基序。另外,在這些基序上游(基序的N端)的7,8或9殘基位置所述基序前面有半胱氨酸殘基(McReynolds et al.(1993)Parasitology today 9 403-406),該殘基的作用在于可以使得蛋白酶的剪切位點遠離該蛋白質(zhì)功能域的主體。
某些寄生蟲的多蛋白(例如犬惡絲蟲的DiAg蛋白)是很強的免疫刺激劑,能夠產(chǎn)生抗原非特異性IgE,并通過干擾寄生蟲特異IgE的產(chǎn)生,在寄生蟲引起的免疫反應(yīng)規(guī)避中起到重要作用。
產(chǎn)生對眾多傳染性疾病起到預(yù)防保護作用的疫苗,是近代醫(yī)學中最重要的進步之一。許多預(yù)防和治療包括自身免疫性、神經(jīng)退行性疾病以及癌癥的其它種類疾病的疫苗目前尚在研發(fā)中。本發(fā)明也可以應(yīng)用于這些其它種類的疾病。在多數(shù)情況下,預(yù)防傳染性疾病的疫苗來自于引起該疾病的介質(zhì)(病原體)的滅活或減毒形式,為了預(yù)防病原體天然形式所引起的感染,可對受體進行該疫苗的注射或其它形式的給藥。受體個體可以通過產(chǎn)生體液(抗體)反應(yīng),細胞(例如細胞溶解性T細胞,CTL)反應(yīng)或兩者同時反應(yīng)來實現(xiàn)免疫反應(yīng)。
對所謂亞單位疫苗(其中的免疫原被定義為傳染性介質(zhì)(agent)或腫瘤抗原的分子片段或亞單位)的開發(fā)一直都是眾多醫(yī)學研究關(guān)注的焦點。用于亞單位疫苗的開發(fā)中的確定候選分子(例如,蛋白或多糖)的需求最初是基于提高安全性的需要,以及在針對細菌性感染的疫苗時,抗菌素耐受問題的增加也是一種原因。然而,與基于整個有機體獲得的疫苗相比,亞單位疫苗的免疫原性越低,而為了產(chǎn)生有效的免疫反應(yīng)(或產(chǎn)生有效的抗腫瘤免疫反應(yīng))以保護目標有機體不受感染,亞單位疫苗對“佐劑”的依賴性也越強。
我們描述了一個蛋白家族,它們可以作為免疫佐劑提高針對于多種預(yù)防性和治療型疫苗所產(chǎn)生的免疫反應(yīng)。佐劑系統(tǒng)在刺激疫苗抗原產(chǎn)生免疫反應(yīng)方面相當有效。新的佐劑系統(tǒng)尤其適用于亞單位疫苗,但對其他種類的疫苗也同樣適用(包括基于整個有機體、核酸等所獲得的疫苗)。
已知線蟲的多梯狀蛋白對誘導(dǎo)IgE反應(yīng)高度有效(Tomlinson et al.J.Immunol.143 2349-2356(1989);Paxton et al.Infect Immunol.612827-2833(1993)。然而,一些多梯狀蛋白(例如犬惡絲蟲的DiAg蛋白)則可以通過產(chǎn)生抗原非特異的IgE來擾亂正常的免疫反應(yīng),其不能與DiAg或寄生蟲結(jié)合(Tezuka,H et al.Infection and Immunity,July 2003,3802-3811),并通過不相關(guān)IgE將寄生蟲特異性的IgE排除在外,來干擾激活的肥大細胞和嗜伊紅細胞對寄生蟲去除。而且,因為IgE抗體常常與危險甚至是致命的過敏反應(yīng)相關(guān)聯(lián)(例如,如果在個體中含有預(yù)先存在的對于所述抗原特異的IgE抗體,該個體與該抗原相遇時就會發(fā)生過敏反應(yīng)),因此IgE反應(yīng)一般被認為是疫苗接種的非理想表現(xiàn)(至少是在針對不是寄生蟲的介質(zhì)的疫苗的情況下)。不僅如此,如果將其作為疫苗物質(zhì),那么可存在多梯狀蛋白增強人個體內(nèi)正在進行中的致敏原特異性的IgE反應(yīng)、或干擾對于寄生蟲預(yù)期的免疫反應(yīng)的有限危險性??傊?,通過多梯狀蛋白誘導(dǎo)產(chǎn)生的IgE反應(yīng)和可以干擾寄生蟲去除或加重過敏性疾病的非特異性IgE反應(yīng),都是將寄生蟲多梯狀蛋白作為疫苗組分或是佐劑進行使用時的禁忌癥。
令人驚奇的是,我們公開了在沒有強烈非特異性IgE反應(yīng)出現(xiàn)的時候,通過例如多梯狀蛋白(或者優(yōu)選與這些蛋白的單重復(fù)單元)的軛合或微粒共制劑的物理性連接是一種可較強地提高針對相聯(lián)抗原產(chǎn)生免疫反應(yīng)的方法。我們公開了多梯狀蛋白(或者優(yōu)選多梯狀蛋白的個體單一功能域部分)和預(yù)期免疫反應(yīng)所針對的抗原的物理性連接,能夠?qū)τ谒龆嗵轄畹鞍椎臐撛谖kU的非抗原特異性IgE反應(yīng)轉(zhuǎn)變?yōu)橛幸娴淖魟┬?yīng),來產(chǎn)生針對所述抗原的預(yù)期抗原特異性抗體。我們還公開了所產(chǎn)生的抗原特異性免疫反應(yīng)(針對多梯狀-功能域-連接的疫苗抗原)是如何傾向于IgG的產(chǎn)生(適用于細菌病原體的去除示例),并傾向于適合去除諸如病毒(和某些寄生蟲)的細胞內(nèi)寄生物的Th1型T細胞反應(yīng)的產(chǎn)生,而不是傾向于有潛在危險的IgE反應(yīng)。我們還公開了多梯狀蛋白或蛋白功能域是如何被用來產(chǎn)生針對相聯(lián)疫苗抗原甚至是當該抗原以非微粒形式進行給藥時的細胞溶解性T淋巴細胞(CTL)反應(yīng)的。除此之外,我們還公開了多梯狀蛋白功能域(甚至是DiAg多梯狀蛋白的功能域)是如何被用來產(chǎn)生針對與其進行物理性連接的抗原(例如寄生蟲抗原而并非DiAg和多梯狀蛋白)的預(yù)期抗原特異性IgE反應(yīng)。
DiAg及其相關(guān)蛋白已被證明在小鼠中能夠?qū)⒚庖呦到y(tǒng)偏離引起自身免疫I型(胰島素依賴型)糖尿病的致病性Th1反應(yīng),并且提倡治療性用于Th1為基礎(chǔ)的自身免疫疾病的治療(Imai,S.et al.Biochem.Biophys.Res.Comm.2861051-1058)。因此令人驚奇的是,在本發(fā)明的另一方面,我們現(xiàn)已公開了DiAg及其相關(guān)蛋白是如何進行過敏性疾病治療的,它們與涉及過敏性疾病的發(fā)病機理中T細胞的作用(Th1)正好完全相反(Th2)。以下對產(chǎn)生DiAg蛋白這種新用途的必需組合物和方法進行了描述。
一方面,本發(fā)明提供作為免疫佐劑用途的多肽,所述多肽包括i)由RXK/RR氨基酸殘基組成的氨基酸基序,R為精氨酸,X為任一氨基酸殘基,及K為賴氨酸;和/或ii)由RXFR氨基酸殘基組成的氨基酸基序,其中F為苯丙氨酸,且在所述基序氨基端前約7-9殘基位置所述基序前面有半胱氨酸殘基,所述多肽能夠通過至少一個氨基酸殘基的添加、缺失或取代來修飾。
一方面,本發(fā)明提供的含有由核酸分子編碼的多肽的佐劑,其中有至少一個RX(K/R)R(其中R為精氨酸,X為任一氨基酸,K/R為賴氨酸或精氨酸,R為精氨酸)或RXFR(其中F為苯丙氨酸)氨基酸基序,在此RX(K/R)R或RXFR基序N端前7,8或9殘基位置該基序前面有半胱氨酸殘基。
優(yōu)選的所述多肽由線蟲的核酸分子編碼。
在本發(fā)明的另一實施方案中,提供了所述蛋白片段的佐劑,并優(yōu)選為淋巴細胞結(jié)合片段。
在本發(fā)明的另一實施方案中,提供了與所述蛋白或與所述蛋白片段、并優(yōu)選淋巴細胞結(jié)合片段具有至少70%同源性的佐劑,。
另一方面,本發(fā)明提供含有至少一種多肽的疫苗組合物,其中所述多肽包括i)由氨基酸殘基RX(K/R)R組成的氨基酸基序,其中R為精氨酸,X為任一氨基酸,K為賴氨酸;和/或ii)由RXFR氨基酸殘基組成的氨基酸基序,其中F為苯丙氨酸,且在所述基序氨基末端前約7-9殘基位置所述基序前面有半胱氨酸殘基,所述多肽能夠通過至少一個氨基酸殘基的添加、缺失或取代來修飾,以及至少一種可以引發(fā)預(yù)期免疫反應(yīng)的抗原來修飾。
在本發(fā)明的優(yōu)選實施方案中,所述多肽與所述抗原混合。
在本發(fā)明的另一優(yōu)選實施方案中,所述多肽與所述抗原軛合,連接或交聯(lián)。
在本發(fā)明的另一優(yōu)選實施方案中,該多肽含有犬惡絲蟲(Dirofilariaimmitis)蛋白,嗜中性白細胞趨化因子(NCF),或其淋巴細胞結(jié)合片段,或其同源結(jié)構(gòu),或其同源結(jié)構(gòu)的淋巴細胞結(jié)合片段。
在本發(fā)明的優(yōu)選實施方案中,所述多肽包括選自SEQ NO.1;2;3;4;5;6;7;8;9;10;11;12;13;14;15;16;17;18;19中的氨基酸序列,或與所選自的序列具有至少50%同源性、更優(yōu)選具有70%的同源性、甚至更優(yōu)選具有90%同源性的多肽。
在本發(fā)明的優(yōu)選實施方案中,所述多肽的長度為至少20個連續(xù)的氨基酸,其序列相同于選自SEQ ID No1;2;3;4;5;6;7;8;9;10;11;12;13;14;15;16;17;18或19序列中的至少20個氨基酸部分。

優(yōu)選的所述核酸序列與所選自的序列具有至少50%的同源性,或更優(yōu)選與所選自的核酸序列具有至少70%的同源性,或更優(yōu)選的與所選自的序列具有至少85%的同源性。
在本發(fā)明的另一實施方案中,所述多肽由選自SEQ ID No20;21;22;23;24;25;26;27;28;29;30;31;32;33或34中的核酸序列的60個核苷酸部分所編碼。
在本發(fā)明的另一實施方案中,佐劑蛋白由與線蟲梯狀蛋白部分同源的蛋白組成,該蛋白部分含有但不包括全部的RX(K/R)R或RXFR序列,并最優(yōu)選避開該序列中的R,K(有時為F)殘基。

這種突變序列的應(yīng)用例子是其中的R和K殘基被甘氨酸殘基“G”所取代的序列。第二個例子是其中R和K殘基被絲氨酸殘基“S”所取代的序列。第三個例子是其中R和K殘基被任意置換的G或S所取代(例如GXGS,SXSG,GXSG等)。
可用的連接序列可能比天然多梯狀蛋白中的長(例如至多達30個殘基),最優(yōu)選5-20個氨基酸殘基,且通常缺乏較強的形成二級結(jié)構(gòu)(例如形成螺旋或形成β-折疊)的傾向以及通常缺乏可被胰蛋白酶樣酶剪切的殘基(原則上是K和R)。
這種形成二級結(jié)構(gòu)傾向的缺乏可用于間隔地連接脯氨酸殘基的工程化,該間隔例如每第三個殘基或者每第十個殘基,更優(yōu)選每第4個,第5個或第6個殘基??蛇x擇地所述脯氨酸可隨機的分布于融合蛋白序列的融合區(qū)域中,使得5個殘基的較小的連接序列含有1個脯氨酸,而15個殘基的較大的連接序列中含有3個或4個脯氨酸。
連接序列的目的在于首先將融合蛋白(也就是多梯狀蛋白半族和抗原半族)中的兩個蛋白部分,以一種對蛋白酶(與天然的多梯狀蛋白序列情況不同,其中所述的功能域間邊界對蛋白酶是非常穩(wěn)定的)相對穩(wěn)定的方式,連接在一起。所述序列的第二個功能是允許蛋白部分在生物合成過程中發(fā)生折疊以形成它們功能屬性(佐劑性,抗原性/免疫原性)所依賴的天然結(jié)構(gòu)域。當使用的連接多肽采用多種形式時,所述連接中不能含有來源于人自身抗原(或欲免疫動物的自身抗原)的序列是非常重要的。因此,通常應(yīng)在數(shù)據(jù)庫中篩選假定的連接序列,以確保連接序列與人類蛋白不具有顯著的同源性,尤其是已知蛋白,或那些懷疑在自身免疫疾病病原學中起重要作用的諸如谷氨酸脫羧酶、胰島素、甲狀腺球蛋白、甲狀腺過氧化物酶、胰島細胞自身抗原、壁細胞自身抗原、腎自身抗原、髓鞘基礎(chǔ)蛋白、髓鞘相關(guān)糖蛋白、髓鞘少突膠質(zhì)糖蛋白的蛋白。
這項普遍規(guī)則的一個例外情況是當這些蛋白以非器官特異性分布于機體,或高豐度表達-例如血液白蛋白和免疫球蛋白的時候。例如,IgG的鉸鏈區(qū)因為其并不是蛋白酶特異不穩(wěn)定的,因此可為所述的融合蛋白提供很好的連接序列。類似地,盡管此序列暴露在外(以IgA的三維結(jié)構(gòu)存在)且易于彎曲,由于僅有少數(shù)幾種蛋白酶(例如腦膜炎球菌IgA蛋白酶)可以剪切這段序列,所以IgA的鉸鏈區(qū)可為所述融合蛋白提供很好的連接序列。由于作為佐劑使用的蛋白開始于一個剪切基序的下游(羧基端),并中止在下一個基序的上游(氨基端),所以RX(K/R)R或RXFR基序可不包括在佐劑蛋白中。佐劑蛋白的起點和終點也可以位于蛋白酶基序的內(nèi)部。
在本發(fā)明的一個優(yōu)選實施方案中,所述多肽可與所述抗原可通過諸如戊二醛或EDC(乙基碳化二亞胺-一種水溶性的碳化二亞胺)的蛋白交聯(lián)劑或優(yōu)選通過諸如MBS或文獻中以及Pierce Chemical Company ofRockford,Illinois,USA或Molecular Probes Inc.of Eugene,Oregon,USA目錄中所述的其他類型的異型雙功能試劑進行軛合或交聯(lián)。
在本發(fā)明的另一優(yōu)選實施方案中,采用本領(lǐng)域中公知的體外DNA重組技術(shù)和克隆技術(shù)方法,通過將編碼抗原和佐劑的核酸進行讀碼框融合,產(chǎn)生含有所述抗原的融合蛋白形式的所述佐劑。
在本發(fā)明的另一優(yōu)選實施方案中,采用合成的微?;蚣{米微粒(例如聚交酯-乙交酯或“PLG”),脂質(zhì)體或免疫刺激復(fù)合物(ISCOMs)將所述多肽和所述抗原包被起來。
微粒制劑是所期望的,因為它引導(dǎo)抗原至抗原提呈細胞,該細胞偏好針對所述抗原的Th1形式的免疫反應(yīng),而且對抗所述多梯狀蛋白部分朝向Th2形式的表現(xiàn)和IgE產(chǎn)生的任何傾向。這種制劑形式在針對抗原產(chǎn)生的所期望細胞介導(dǎo)的和IgG抗體反應(yīng)的刺激是有用的。微粒制劑還能容易地整合用來將免疫反應(yīng)偏移至Th1方向上的附加材料。這些材料通常包括抗IL10和IL4,諸如IFN-γ的Th1細胞因子和CpG DNA的抗體。
最好的情況是,微粒制劑包含所述的多梯狀蛋白部分和抗原,針對該抗原在同一個微粒中形成預(yù)期的免疫反應(yīng),從而使制劑中的每一個微粒都有效負載地攜帶兩個實體。這種制劑可以保證兩種物質(zhì)被任何給定的單一抗原體呈細胞攝取并最大化對于所述有效負載抗原的微粒制劑的Th1傾向作用,甚至當此抗原和多梯狀功能域(以及上一段中描述的任意的Th1傾向性物質(zhì))并沒有相連時,同時存在于同一微粒中除外。
微粒制劑優(yōu)選具有顯著程度的多梯狀蛋白部分和抗原部分的表面暴露(5-10%)。一般情況下,微粒制劑過程中就可默認形成這種暴露程度。如果這樣的暴露程度不能實現(xiàn),上述蛋白部分可以通過共價軛合的形式軛合到所述微粒表面??乖糠值谋砻姹┞队欣诖碳た乖禺愋訠細胞,并且有助于對于抗原的抗體反應(yīng)。
通常這些微粒的大小為150納米最高至10微米直徑。更優(yōu)選它們?yōu)?00納米最高至2微米。
在本發(fā)明的一個優(yōu)選實施方案中,所述多肽和抗原共吸收或共沉淀在諸如氫氧化鋁凝膠或磷酸鈣的鋁或鈣鹽上。

優(yōu)選的所述載體是一種“穿梭載體”,可以在大腸桿菌中進行擴增,并通過適合的諸如CMV或其它真核啟動子的啟動子在哺乳動物細胞中表達融合蛋白。
根據(jù)本發(fā)明另一方面,佐劑蛋白功能域(例如來自于犬惡絲蟲的NCF)可由幾個拷貝(最優(yōu)選1,2或3拷貝)代表,并作為同一條多肽鏈的部分與抗原(單一拷貝)形成共線性融合。可選擇地,佐劑蛋白功能域以單拷貝存在,與抗原蛋白融合形成寡聚體(例如二聚體,三聚體,四聚體等)。在后者的構(gòu)建中,一旦抗原蛋白發(fā)生寡聚化,佐劑蛋白功能域就成為寡聚結(jié)構(gòu)。最方便的情況是,單拷貝的佐劑蛋白與來自于傳染性介質(zhì)的寡聚蛋白形成讀碼框融合,針對該介質(zhì)進行預(yù)防疫苗設(shè)計,例如流感紅細胞凝集素或HIV衣殼糖蛋白gp120。可選擇地,單拷貝佐劑蛋白功能域與不發(fā)生寡聚化的抗原相融合。在這種情況下,寡聚體形式可以通過與具有天然寡聚化傾向的蛋白部分(例如卷曲化卷曲)結(jié)合來產(chǎn)生。適合的寡聚化部分的例子為鏈球菌中(例如M蛋白)配對的螺旋卷曲化卷曲結(jié)構(gòu),其可形成二聚體卷曲化卷曲;同樣也可以使用三聚體螺旋蛋白部分。其中一個例子是諸如CD23的2型膜蛋白的莖結(jié)構(gòu)。被設(shè)計用來研究卷曲化卷曲蛋白的聚合性質(zhì)的人工卷曲化卷曲多肽也是合適的。最優(yōu)選的寡聚化程度是三聚體,因為它反應(yīng)出犬惡絲蟲(D.immitis)蛋白的預(yù)測天然存在形式。在佐劑功能域以多個拷貝的形式出現(xiàn)的情況下,最優(yōu)選的實施方案將是犬惡絲蟲蛋白的天然重復(fù)結(jié)構(gòu)。當結(jié)構(gòu)域作為單一多肽鏈的部分進行重復(fù)時,最優(yōu)選的是在DNA水平上發(fā)掘可替代的密碼子的使用,以避免直接的DNA序列重復(fù),該序列在編碼DNA擴增過程中會另外產(chǎn)生同源重組的問題和缺失問題。
通過采用交聯(lián)劑將佐劑和抗原相連以形成軛合物。可選擇地軛合物也可以是佐劑和抗原的翻譯融合物。

啟動子是本領(lǐng)域認知的術(shù)語,為了清晰地對其進行說明,其包括以下僅由例子提供的特征,并不是任何方式的限制。增強子元件是一段通常存在于基因(增強子也可以存在于基因序列的3’端,甚至位于內(nèi)含子序列的內(nèi)部,因此它們是位置獨立的)轉(zhuǎn)錄起始位點5’端的順式作用核酸序列。增強子的功能是增加與其相連基因的轉(zhuǎn)錄效率。增強子的活化依賴于與增強子元件特異性結(jié)合的反式作用轉(zhuǎn)錄因子(多肽)。轉(zhuǎn)錄因子的結(jié)合/活化(詳見Eukaryotic Transcription Factors,by David S Latchman,Academic Press Ltd,San Diego)與很多環(huán)境相關(guān)因素有關(guān),其包括例如而并非為對其進行限制,中間代謝產(chǎn)物、環(huán)境效應(yīng)因子。

材料與方法1.抗原-佐劑軛合物的制備a.重組佐劑蛋白的制備采用多聚酶鏈式反應(yīng)(PCR),利用引物(5_引物,包含NdeI限制性內(nèi)切酶識別位點5_GCATATGAATGATCATAATTTAGAAAGC-3,3_引物,包含BamHI限制性內(nèi)切酶識別位點5_-CTAAAGGATCCTATCACCGCTTACGCCGTTCATTCATTG-3),從犬惡絲蟲的cDNA文庫對DiAg基因的V1功能域(一個重復(fù)單元)進行擴增。用NdeI和BamHI消化擴增的DNA后克隆到pET3a載體上(Stratagene),在大腸桿菌HMS174(DE3)中進行表達。根據(jù)以下所述進行rDiAg的純化。用30ml 50mM HCl和5mMEDTA在4℃重懸5g細胞團,然后12,000 g離心10min。采用60-80%的飽和硫酸銨對上清液中的重組DiAg進行沉淀后過Superdex 200柱。采用固定化的多粘菌素B移去濃縮rDiAg溶液中大腸桿菌熱源污染物。純化后的佐劑蛋白經(jīng)過凍干處理后儲存于-20℃直至使用。
b.佐劑蛋白與抗原的軛合下面僅對從多種將抗體和多肽偶聯(lián)的方法選出的一種作為示例進行描述將犬惡絲蟲多梯狀蛋白V1結(jié)構(gòu)域佐劑(YFQTYLSWLTDAQKDEIKKMKEEGKSKMIQKKIFDYFESLTGDKKKKAAEELQQGCLMALSEIIGNEKMLMLKEIKDSGADPEQIEDMLKLVVDKEKKKRIDEYPPVCRKIYAAMNERRK)以3-30mg/ml的濃度溶解于pH 7.2的0.1M磷酸鈉、0.15M NaCl溶液中。向其中加入6mg的硫代-SMCC(Pierce),將該混合物于室溫孵育30min。采用pH 7.2的0.1M磷酸鈉,0.15M NaCl作為層析緩沖液,以脫鹽柱(Sephadex G-25)或超濾的方法可隨即去除過量的交聯(lián)物。將含有多肽(經(jīng)OD280測定)的組分合并,并將馬來酰亞胺激活的多肽濃縮到大約10mg/ml。
與此同時,抗原例如,純化的重組HSV糖蛋白D抗原(gD)以1-5mg/ml的濃度溶解或交換于pH 7.2的0.1M磷酸鈉、0.15M NaCl溶液中。以為1mg/ml濃度,在每ml gD中加入10-40μl SATA(Pierce)儲存液(8mg/ml溶于DMSO或DMF)。室溫反應(yīng)30min。通過透析、凝膠過濾或超濾從未反應(yīng)的SATA中純化gD。
位于SATA修飾gD上的乙酰巰基化基團通過以下描述進行脫保護制備pH7.2的含有10mM EDTA的0.1M磷酸鈉中的0.5M羥胺溶液。每毫升抗體中加入100μl上述溶液,室溫放置2小時。將硫醇化的gD超濾純化到pH7.2的0.1M磷酸鈉、0.1M氯化鈉、10mM EDTA溶液,并隨即與經(jīng)馬來酰亞胺處理的DiAg以gD∶DiAg的1∶10的摩爾比混合。37C反應(yīng)持續(xù)兩小時。采用超濾的方法從未發(fā)生反應(yīng)的DiAg中純化軛合的gD-DiAg。
2.抗原佐劑融合蛋白的制備采用多聚酶鏈式反應(yīng)(PCR),利用引物(5_引物,包含HindII限制性內(nèi)切酶識別位點5_GAAGCTTAATGATCATAATTTAGAAAGC-3,3_引物,包含BamHI限制性內(nèi)切酶識別位點5_-CTAAAGGATCCTATCACCGCTTACGCCGTTCATTCATTG-3),從犬惡絲蟲的cDNA文庫擴增犬惡絲蟲多梯狀蛋白的V1功能域(一個重復(fù)單元)。用HindIII和BamHI消化擴增的DNA。
與此同時,從HSV感染的細胞,使用結(jié)合附加的核酸的引物和在5’引物具有BamHI識別序列的引物,通過擴增得到gD編碼DNA,從而通過BamHI消化并與DiAg的編碼片段相連,使編碼DiAg和gD的cDNA是相互在共同的讀碼框中,使得所述DNA連續(xù)地轉(zhuǎn)錄為可翻譯成由DiAg和gD組成的融合蛋白的mRNA。將這兩個DNA片段連入哺乳動物表達載體(pcDNA3.1(Invitrogen)),并將該質(zhì)粒轉(zhuǎn)化感受態(tài)大腸桿菌以產(chǎn)生足量的質(zhì)粒(使用Qiagen柱進行純化)來對COS-7細胞進行電穿孔轉(zhuǎn)染。3-5天后,采用抗-DiAg親和柱對轉(zhuǎn)染的COS-7細胞的上清液進行純化,在經(jīng)過凝膠過濾的進一步純化后,融合蛋白就可以用來進行免疫了。
3.編碼抗原-佐劑融合蛋白的DNA疫苗的制備含有插入片段并如上所述制備的pcDNA3.1表達載體,可以直接作為DNA疫苗通過肌肉注射來使用。
