本發(fā)明涉及基因工程及農(nóng)業(yè)類畜牧獸醫(yī)領(lǐng)域,具體地說,涉及一種利用乳腺生物反應(yīng)器提高乳汁中褪黑素含量的方法。
背景技術(shù):
褪黑素(n-acetyl-5-methoxytryptamine)是一種生物鐘的調(diào)節(jié)激素,參與神經(jīng)和外周組織細(xì)胞時(shí)鐘基因的表達(dá)調(diào)控。褪黑素是兩親分子,容易進(jìn)入所有的亞細(xì)胞結(jié)構(gòu)空間,包括細(xì)胞膜膜結(jié)構(gòu)、細(xì)胞質(zhì)、細(xì)胞核和線粒體中。在各種各樣復(fù)雜的生理活動中,褪黑素是一種強(qiáng)效的活性氧(ros)和活性氮(rns)的自由基清除劑。乙?;D(zhuǎn)移酶(aanat)和羥基吲哚-o-甲基轉(zhuǎn)移酶(hiomt)被證明是合成褪黑素的關(guān)鍵限速酶。血液褪黑素主要合成器官在松果體中,5-羥色胺(5-ht)來自血液中的色氨酸的脫羧反應(yīng),在aanat的催化下轉(zhuǎn)變?yōu)閚-乙酰基-五羥色胺(nas);在乙酰血清素-o-甲基轉(zhuǎn)移酶(hiomt)的催化下,合成褪黑素。松果體中aanat酶的活力受光的抑制,表現(xiàn)出晝夜節(jié)律,其活力與血液及松果體褪黑素水平一致;hiomt在褪黑素周期中發(fā)揮重要作用,其活力與褪黑素的產(chǎn)生呈正相關(guān)。研究表明這兩種酶是褪黑素合成途徑中的重要催化劑,直接影響褪黑素的合成產(chǎn)量。
褪黑素在不同組織中具有多種生物活性。褪黑素的代謝產(chǎn)物n-acetyl-n-formyl-5-methoxykynuramine(afmk)和n-acetyl-5-methoxykynuramine(amk)也是高效的抗氧化劑。褪黑素與自由基發(fā)生的反應(yīng)是一種不可逆的氧化反應(yīng),不能被循環(huán)利用,它在氧化過程中形成的各級中間代謝產(chǎn)物都具有顯著的抗氧化能力,有的作用甚至超過褪黑素本身,形成褪黑素級聯(lián)放大的抗氧化機(jī)制。褪黑素被認(rèn)為是一種抗衰老的激素,它還具有提高睡眠質(zhì)量和發(fā)展免疫的功能。其中,已知褪黑素可以影響胃腸道平滑肌,具有放松平滑肌和催眠的效果。它可能有助于嬰兒更好的夜間睡眠,從而促進(jìn)免疫系統(tǒng)的發(fā)展和早期的晝夜節(jié)律建立。外源低濃度的褪黑素可以迅速吸收,發(fā)揮抗氧化、抗炎癥作用從而影響并調(diào)節(jié)免疫功能。例如,國外已有高褪黑素含量牛奶“nightmilk”等上市,有更好的促進(jìn)睡眠效果和提高免疫力的功能。在綿羊乳腺中高表達(dá)褪黑素合成限速酶aanat和hiomt,催化5-羥色胺合成高水平褪黑素,使乳汁中褪黑素含量進(jìn)一步提高,不僅可以使羔羊增強(qiáng)免疫力,提高成活率;同時(shí)對于褪黑素功能乳的開發(fā)奠定基礎(chǔ),提高東弗利生等乳用羊的產(chǎn)品附加值。
技術(shù)實(shí)現(xiàn)要素:
本發(fā)明的目的是提供一種利用乳腺生物反應(yīng)器提高乳汁中褪黑素含量的方法。
為了實(shí)現(xiàn)本發(fā)明目的,本發(fā)明首先提供過表達(dá)褪黑素關(guān)鍵合成酶aanat和hiomt基因的原核胚的構(gòu)建方法,包括以下步驟:
(1)分別將褪黑素關(guān)鍵合成酶aanat和hiomt的cdna插入表達(dá)載體中,構(gòu)建得到重組表達(dá)載體;
(2)cas9cdna及sgrna的制備:將經(jīng)過密碼子優(yōu)化后的cas9序列及sgrna序列分別構(gòu)建到包含t7啟動子的載體上,然后通過體外轉(zhuǎn)錄分別得到cas9cdna及sgrna產(chǎn)物并純化;
其中,sgrna的作用位點(diǎn)位于羊mstn基因第三外顯子區(qū),sgrna作用位點(diǎn)的核苷酸序列為5’-gacatctttgtaggagtacagcaa-3’;
cas9cdna的核苷酸序列如seqidno:1所示;
(3)將(1)構(gòu)建的重組表達(dá)載體線性化,然后與cas9cdna及sgrna一起導(dǎo)入哺乳動物的原核期胚胎細(xì)胞中,即得。
本發(fā)明所述褪黑素關(guān)鍵合成酶aanat和hiomt的cdna來自羊松果體細(xì)胞。優(yōu)選杜泊羊松果體細(xì)胞。
前述方法中,步驟(1)所述表達(dá)載體為pbc1(攜帶有酪蛋白啟動子)。分別將aanat和hiomt的cdna插入載體pbc1的xhoi多克隆位點(diǎn)之間,得到重組表達(dá)載體pbc1-aanat和pbc1-hiomt(質(zhì)粒結(jié)構(gòu)見圖9和圖10,載體pbc1-aanat的全序列如seqidno:2所示,將seqidno:2中第8614~9240位堿基替換成seqidno:3所示序列即為載體pbc1-hiomt的全序列)。
前述的方法,步驟(3)是將重組表達(dá)載體pbc1-aanat、pbc1-hiomt分別用sali和noti限性內(nèi)切酶進(jìn)行線性化處理,分別回收大片段(長度分別為16391bp和16795bp),然后與cas9cdna及sgrna一起通過原核顯微注射直接注入羊原核胚的細(xì)胞質(zhì)中。
進(jìn)行顯微注射時(shí),大片段的注射濃度為80ng/μl;cas9cdna的注射濃度為838ng/μl;sgrna的注射濃度為38.7ng/μl;單枚原核胚注射總體積為10pl。
本發(fā)明還提供按照上述方法制備的過表達(dá)褪黑素關(guān)鍵合成酶aanat和hiomt基因的原核胚。
本發(fā)明還提供所述原核胚在制備動物乳腺反應(yīng)器、轉(zhuǎn)基因動物以及轉(zhuǎn)基因細(xì)胞系中的應(yīng)用。
本發(fā)明還提供一種利用乳腺生物反應(yīng)器提高乳汁中褪黑素含量的方法,其是將所述原核胚移入受體羊的輸卵管中,經(jīng)過妊娠、分娩,獲得陽性轉(zhuǎn)基因羊,獲得的轉(zhuǎn)基因羊的原代或后代母羊在泌乳期的乳汁中產(chǎn)生褪黑素,從而提高乳汁中褪黑素含量。
其中,用于鑒定陽性轉(zhuǎn)基因羊的特異性pcr引物包括:

所述泌乳期包括人工誘導(dǎo)泌乳期和自然分泌泌乳期;所述人工誘導(dǎo)泌乳期和自然泌乳期包括泌乳前期、泌乳中期和泌乳后期。
本發(fā)明所述羊?yàn)榫d羊。
本發(fā)明進(jìn)一步提供按照上述方法制備的褪黑素功能乳。
本發(fā)明具有以下優(yōu)點(diǎn):
(一)本發(fā)明利用羊乳腺反應(yīng)器合成褪黑素限速酶,從而生產(chǎn)天然褪黑素和褪黑素功能乳。在將羊自身的褪黑素合成酶cdna連接酪蛋白啟動子整合到羊基因組中,最大程度地減少了轉(zhuǎn)基因動物自身的外來蛋白的影響。轉(zhuǎn)入的基因中無標(biāo)記、無抗性基因、也無細(xì)菌載體片段,不僅避免了密碼子偏好的問題,更符合轉(zhuǎn)基因安全的要求。
(二)通過高效的crispr/cas9系統(tǒng),制作褪黑素乳腺生物反應(yīng)器,獲得的羊aanat和hiomt基因拷貝數(shù)增加1個(gè)以上,aanat和hiomt蛋白表達(dá)上升。利用乳腺生產(chǎn)高水平天然褪黑素,不僅可以提高乳腺健康水平、生產(chǎn)高質(zhì)量的乳汁,增強(qiáng)羔羊免疫力,提高羔羊成活率;還可以解決目前褪黑素主要依賴化學(xué)合成,造成微量雜質(zhì)殘留的問題,又可以規(guī)避生物提取潛在的病原物質(zhì)感染的風(fēng)險(xiǎn)。而且羊乳腺反應(yīng)器和產(chǎn)品均具有重要的研究價(jià)值和商業(yè)前景。
(三)實(shí)驗(yàn)表明,本發(fā)明的陽性轉(zhuǎn)基因羊乳腺細(xì)胞褪黑素合成酶的表達(dá)量顯著提高。在催乳素誘導(dǎo)下,陽性乳腺細(xì)胞可合成高水平褪黑素,并且陽性羊血液中的褪黑素水平也顯著高于普通羊。轉(zhuǎn)基因羊整個(gè)泌乳期階段都具有分泌褪黑素合成酶的能力,并且通過育種選擇,能進(jìn)一步提高轉(zhuǎn)基因羊的褪黑素表達(dá)量。

具體實(shí)施方式
以下實(shí)施例用于說明本發(fā)明,但不用來限制本發(fā)明的范圍。若未特別指明,實(shí)施例均按照常規(guī)實(shí)驗(yàn)條件,如sambrook等分子克隆實(shí)驗(yàn)手冊(sambrookj&russelldw,molecularcloning:alaboratorymanual,2001),或按照制造廠商說明書建議的條件。
實(shí)施例1利用羊乳腺生物反應(yīng)器提高乳汁中褪黑素含量的方法
1.基因克隆和表達(dá)載體構(gòu)建
從杜泊羊松果體中提取總rna(ovisaries),合成cdna第一鏈。分別擴(kuò)增出aanat和hiomt完整的開放閱讀框(orf)和部分上下游非編碼區(qū)(引物見表1)。aanat和hiomt的pcr產(chǎn)物單酶切后連接到pmdtm19-t載體中測序,基因片段的大小分別為634bp和1048bp。測序驗(yàn)證后的cdna片段連接到乳腺特異性表達(dá)載體pbc1(invitrogen)上。重組載體命名為pbc1-aanat和pbc1-hiomt(seqidno:1和2,質(zhì)粒結(jié)構(gòu)見圖9和圖10)。兩者均由β-酪蛋白啟動子驅(qū)動生產(chǎn)重組蛋白。線性化pbc1-aanat/hiomt載體結(jié)構(gòu)見圖1。

2.cas9cdna及sgrna的制備
將經(jīng)過密碼子優(yōu)化后的cas9序列及sgrna序列分別構(gòu)建到包含t7啟動子的載體pcdna3.1(+)(invitrogen)上,然后通過體外轉(zhuǎn)錄分別得到cas9cdna及sgrna產(chǎn)物并純化。
其中,sgrna的作用位點(diǎn)位于羊mstn基因第三外顯子區(qū),sgrna作用位點(diǎn)的核苷酸序列為5’-gacatctttgtaggagtacagcaa-3’。
cas9cdna的核苷酸序列如seqidno:1所示。
3.原核胚細(xì)胞質(zhì)內(nèi)顯微注射生產(chǎn)轉(zhuǎn)基因羊
乳腺表達(dá)載體pbc1-aanat和pbc1-hiomt進(jìn)行線性化處理。利用sali和noti限性內(nèi)切酶可以釋放5.9kb的細(xì)菌質(zhì)粒部分與線性dna大片段,dna大片段長度分別為16391bp和16795bp。線性dna為表達(dá)構(gòu)件、包含調(diào)控元件和插入的目的基因cdna。
以杜泊羊?yàn)楣w進(jìn)行超數(shù)排卵。通過等量混合pbc1-aanat和pbc1-hiomt兩個(gè)表達(dá)載體,以及cas9cdna和sgrna,采用原核顯微注射直接注入羊原核胚的細(xì)胞質(zhì)中,生產(chǎn)過表達(dá)褪黑素合成基因的綿羊后代。
大片段的注射濃度為80ng/μl;cas9cdna的注射濃度為838ng/μl;sgrna的注射濃度為38.7ng/μl;單枚原核胚注射總體積為10pl。
從62只供體羊中共獲得593枚原核注射胚胎,移植到所有150個(gè)受體母羊輸卵管內(nèi),21天超聲檢查后77只妊娠,妊娠率為51.33%(表2)。我們共獲得98只胚胎移植綿羊后代,其中包括經(jīng)測序鑒定的34只轉(zhuǎn)基因陽性羔羊,產(chǎn)羔率和轉(zhuǎn)基因pcr測序鑒定陽性率分別為65.33%和34.69%(表2)。其中,有7只攜帶外源aanat基因,2只攜帶外源hiomt基因,其余25羔羊同時(shí)攜帶aanat和hiomt兩種基因。

*a8-f、h4-f分別鑒定轉(zhuǎn)入的aanat、hiomt基因,與mlt-r共同使用;利用pbc1-f/r鑒定轉(zhuǎn)aanat基因后代,擴(kuò)增產(chǎn)物大小為765bp,利用pbc1-f/r鑒定轉(zhuǎn)hiomt基因后代,擴(kuò)增產(chǎn)物大小為1169bp。
通過southern印跡雜交法進(jìn)一步驗(yàn)證pcr測序結(jié)果(圖2)。使用dig標(biāo)記aanat基因的探針分別與個(gè)體基因組及載體質(zhì)粒雜交?;蚪M與載體質(zhì)粒均用hindⅲ單酶切徹底消化,獲得陽性條帶約為5.2kb。wt為野生型個(gè)體,p為表達(dá)載體。重復(fù)驗(yàn)證個(gè)體共5個(gè),標(biāo)號分別為1-5號陽性個(gè)體。
4.細(xì)胞系中aanat和hiomt基因表達(dá)水平
從整合外源aanat和hiomt基因的母羊乳腺處取得上皮細(xì)胞,經(jīng)催乳素誘導(dǎo)培養(yǎng)基培養(yǎng),以便通過β-酪蛋白啟動子啟動外源基因的表達(dá)。q-pcr檢測發(fā)現(xiàn)在陽性轉(zhuǎn)染細(xì)胞系中,與轉(zhuǎn)染空白載體和對照組細(xì)胞系相比,24h至48h范圍內(nèi)aanat和hiomt基因在mrna水平表達(dá)差異極顯著(n=4,p<0.01;圖3)。

5.轉(zhuǎn)基因后代aanat和hiomt基因表達(dá)水平
獲得轉(zhuǎn)aanat和hiomt雙基因群體后,分別在早晨4:00和下午16:00對其血清中褪黑素水平進(jìn)行了測定。轉(zhuǎn)基因組與野生型組羊相比4:00和16:00褪黑素濃度差異不顯著(n=10,p>0.05)(圖5)。
取泌乳期轉(zhuǎn)基因和野生型羊乳腺上皮細(xì)胞,原代培養(yǎng)下并檢測基因表達(dá)情況。褪黑素的測定表明,通過培養(yǎng)基添加底物5-ht(10-4mol/l),2小時(shí)后發(fā)現(xiàn)野生型和轉(zhuǎn)基因細(xì)胞之間差異顯著(n=4,p<0.01。圖6)。q-pcr顯示轉(zhuǎn)基因個(gè)體與陰性或是野生型個(gè)體相比,mrna轉(zhuǎn)錄水平的差異極顯著(n=4,p<0.01),高出約10倍(圖7)。western印記雜交法測定活性受調(diào)控的合成限速酶aanat表達(dá)水平。轉(zhuǎn)基因合成限速酶aanat(23kd)表達(dá)水平明顯高于野生型(1-4為轉(zhuǎn)基因個(gè)體,4-8為野生型個(gè)體。內(nèi)參actin-β43kd,圖8a)。分別在4:00和16:00對轉(zhuǎn)基因組羊奶中褪黑素水平進(jìn)行了測定。轉(zhuǎn)基因組羊奶中夜間褪黑素含量極顯著地高于野生型組(n=4,p<0.01,圖8b),提高3倍左右。
本發(fā)明利用原核注射技術(shù)生產(chǎn)轉(zhuǎn)基因羊,快速引入新的基因——褪黑素合成限速酶基因,產(chǎn)生期望的生產(chǎn)性狀——乳中高含量天然褪黑素。并在此基礎(chǔ)上建立育種群體,進(jìn)一步選育表達(dá)性狀優(yōu)良的個(gè)體,為家畜新品種培育打下良好基礎(chǔ)。同時(shí)能提高羔羊的健康狀況、提高存活率,為羊養(yǎng)殖產(chǎn)業(yè)服務(wù)。在新品種培育的基礎(chǔ)上,該技術(shù)用于開發(fā)褪黑素功能乳產(chǎn)品,發(fā)揮其保健功能的作用以便人類利用,具有積極的社會效益。
羊奶營養(yǎng)價(jià)值優(yōu)良,經(jīng)濟(jì)效益高,隨著國內(nèi)消費(fèi)水平的提高,羊奶的市場前景非常廣闊。通過乳腺特異表達(dá)載體實(shí)現(xiàn)褪黑素合成酶在綿羊乳腺中高效表達(dá),使乳腺分泌高水平褪黑素,改善母羊乳腺功能和羊奶品質(zhì),進(jìn)一步獲得具有商業(yè)開發(fā)價(jià)值的褪黑素功能乳轉(zhuǎn)基因羊,可帶來巨大的經(jīng)濟(jì)效益。
與傳統(tǒng)的原核顯微注射生產(chǎn)轉(zhuǎn)基因動物的方法(轉(zhuǎn)基因小鼠陽性率約10%,轉(zhuǎn)基因羊陽性率約2~10%)相比,本發(fā)明具有很高的陽性率達(dá)到34.69%。同時(shí),相比傳統(tǒng)的原核注射技術(shù),本方法可實(shí)現(xiàn)外源基因的定點(diǎn)整合,可控性和安全性高。
雖然,上文中已經(jīng)用一般性說明及具體實(shí)施方案對本發(fā)明作了詳盡的描述,但在本發(fā)明基礎(chǔ)上,可以對之做一些修改或改進(jìn),這對本領(lǐng)域技術(shù)人員而言是顯而易見的。因此,在不偏離本發(fā)明精神的基礎(chǔ)上所做的這些修改或改進(jìn),均屬于本發(fā)明要求保護(hù)的范圍。
