本發(fā)明屬于生物和醫(yī)學(xué)實驗?zāi)P偷闹苽浼夹g(shù)和細胞構(gòu)建領(lǐng)域。涉及GLCCI1基因基于Cre-LoxP條件性基因敲除小鼠模型構(gòu)建試劑盒、打靶載體及構(gòu)建方法。
背景技術(shù):
GLCCI1基因,即糖皮質(zhì)激素誘導(dǎo)的轉(zhuǎn)錄基因1,位于第7號染色體短臂的第2區(qū)1號帶的第3亞帶(7p21.3),包含8個外顯子,在全身多種組織器官中均有表達。GLCCI1最早由Chapman等描述,他們發(fā)現(xiàn)對胸腺來源的細胞系進行地塞米松刺激后,在激素敏感型和激素不敏感型的細胞中某些基因序列標(biāo)簽表達存在明顯差異。文獻報道GLCCI1與多種疾病的發(fā)生及激素治療療效有關(guān),包括支氣管哮喘、腎病綜合征等。然而,GLCCI1基因的功能目前尚不明確,構(gòu)建一種GLCCI1基因敲除小鼠模型,將有利于深入開展GLCCI1的功能及其在相關(guān)疾病中的發(fā)病機制研究,為這些疾病的治療提供可能的作用靶點。
基因敲除動物模型是活體動物上開展基因功能研究、尋找作用靶標(biāo)的重要工具。小鼠是研究人類疾病的一種重要模式生物。通過利用Cre-loxP重組系統(tǒng),可以將基因元件引入打靶載體上,實現(xiàn)條件性基因敲除,從而構(gòu)建條件性基因敲除小鼠模型,為研究者提供基因功能研究的良好平臺。
技術(shù)實現(xiàn)要素:
本發(fā)明旨在克服現(xiàn)有技術(shù)的不足,提供GLCCI1基因基于Cre-LoxP條件性基因敲除小鼠模型構(gòu)建試劑盒、打靶載體及構(gòu)建方法。
為了達到上述目的,本發(fā)明提供的技術(shù)方案為:
所述GLCCI1基因基于Cre-LoxP條件性基因敲除小鼠模型構(gòu)建試劑盒中包括如下引物對:
5’端篩選引物對:

所述GLCCI1基因基于Cre-LoxP條件性基因敲除小鼠模型構(gòu)建的打靶載體的序列如SEQ ID NO.13所示,所述打靶載體命名為Glcci1-final vector。
所述GLCCI1基因基于Cre-LoxP條件性基因敲除小鼠模型的構(gòu)建方法包括如下步驟:
(1)構(gòu)建打靶載體Glcci1-final vector;
(2)ES細胞篩選及鑒定;
(3)制備嵌合小鼠;
(4)繁育F1代雜合子小鼠;
(5)獲得GLCCI1-/-純合子小鼠,即為GLCCI1基因基于Cre-LoxP條件性基因敲除小鼠模型。
優(yōu)選地,所述步驟(1)是采用BAC載體構(gòu)建打靶載體Glcci1-final vector,利用含有GLCCI1的BAC載體套取得到GLCCI1的同源重組臂,通過同源重組的方式將1st-loxp插入exon 2的上游,將2nd-loxp插入exon 2的下游,將構(gòu)建好的打靶載體Glcci1-final vector用PCR及ApaLⅠ、BglⅡ、KpnⅠ、PstⅠ酶切鑒定,PCR鑒定的引物為:5’-CTGCTTTGAGGTCTTCATCTGC-3’(SEQ ID NO.5),5’-CTTACAGTGCCTCCTCATTACC-3’(SEQ ID NO.6);。
所述步驟(2)是將打靶載體Glcci1-final vector采用電沖擊法轉(zhuǎn)到ES細胞中進行打靶,使其發(fā)生同源重組,通過PCR及Southern blot篩選出中靶克?。黄渲?,用于5’端篩選的PCR反應(yīng)引物對為:5’-CAGTTCCTGCATTAGCCACCAC-3’(SEQ ID NO.1),5’-CTAGTTGGCTGGACGTAAACT-3’(SEQ ID NO.2);用于3’端篩選的PCR反應(yīng)引物對為:5’-ACAGGACCTTACCTCACAGG-3’(SEQ ID NO.3),5’-CTAGAACTAGTGGATCCCCTCGA-3’(SEQ ID NO.4);優(yōu)選地,5’端篩選的PCR反應(yīng)條件為95℃10秒、68℃6分鐘,共40個循環(huán);3’端篩選的PCR反應(yīng)條件為95℃30秒、58℃30秒,72℃40秒,共35個循環(huán)。
在步驟(3)中,注射用囊胚采用C57BL/6N近交系小鼠,通過超數(shù)排卵,自然受孕,胚胎體內(nèi)發(fā)育至囊胚階段,用于注射;注射后移植入假孕小鼠受體子宮,假孕受體為C57BL/6N與CBA的雜交一代,生育出嵌合率大于50%的嵌合雄鼠。
所述步驟(4)是將獲得的嵌合率高的嵌合雄鼠與野生型C57BL/6N背景雌鼠回交,得到F1代雜合子;經(jīng)提取尾基因組DNA進行PCR反應(yīng)鑒定基因型,根據(jù)電泳結(jié)果判斷野生型、雜合子或純合子。步驟(4)中,用于F1代雜合子小鼠基因型鑒定的PCR反應(yīng)的引物分別為:①檢測1st loxP:GLCCI1-FRT-tF:5’-CTGCTTTGAGGTCTTCATCTGC-3’(SEQ ID NO.7);Neo-5R:5’-GGCTGGACGTAAACTCCTC-3’(SEQ ID NO.8)。②檢測2nd loxP:GLCCI1-loxptF3:5’-CTGGCAGTTTGTGAGGCATG-3’(SEQ ID NO.9);ZMK-1R3:5’-AAGGGTTATTGAATATGATCGGA-3’(SEQ ID NO.10)。③檢測內(nèi)參:GLCCI1-loxp-tF:5’-CCCTTGTTTTACATGCGGAGC-3’(SEQ ID NO.11);GLCCI1-loxp-tR:5’-GCTCACAGTCAGCTAATGGATGG-3’(SEQ ID NO.12)。反應(yīng)條件為:95℃30秒;58℃30秒;72℃30秒;35個循環(huán)。
所述步驟(5)是將獲得的F1代雜合子小鼠與先后與Flper小鼠及野生型C57BL/6N小鼠交配,獲得完全刪去Neo抗性基因的小鼠,再將完全刪去Neo抗性基因的小鼠與全身性敲除cre品系交配獲得全身性敲除GLCCI1的純合子小鼠,即為GLCCI1基因基于Cre-LoxP條件性基因敲除小鼠模型。
本發(fā)明的有益效果在于,通過建立和應(yīng)用GLCCI1基因敲除在整體水平、細胞水平及分子水平深入探討GLCCI1在支氣管哮喘、腎病綜合征等機體疾病發(fā)生、發(fā)展中的作用和機制,并探討GLCCI1與疾病的激素治療療效的關(guān)聯(lián)。
傳統(tǒng)基因打靶小鼠的建系多使用小質(zhì)粒和DNA雜交等方法構(gòu)建和鑒定。本實驗使用BAC載體和PCR加測序鑒定獲得同樣結(jié)果。用同源重組方法敲除ES干細胞的靶基因,經(jīng)過嵌合體、雜交生育繁殖工程和篩選鑒定,獲得GLCCI1基因敲除純合子小鼠。目前關(guān)于GLCCI1在各種疾病中的作用的研究相繼出現(xiàn),但均無法深入探討GLCCI1在疾病中的確切作用,GLCCI1基因敲除小鼠的出現(xiàn),將使GLCCI1的研究獲得實質(zhì)性進展并有利于相關(guān)疾病的發(fā)病機制及治療方案研究。
總之,通過利用Cre/loxP原理,在GLCCI1的一段目標(biāo)序列兩端各放置一個loxP序列,得到flox小鼠,將flox小鼠與全身性敲除Cre品系的小鼠交配繁殖,獲得全身性敲除GLCCI1基因的小鼠品系,獲得的GLCCI1基因敲除小鼠將可應(yīng)用于GLCCI1基因及蛋白質(zhì)功能研究。

具體實施方式
以下通過具體的實施例進行示例,其中所用試劑均可通過市場渠道購買,其它可以參考本領(lǐng)域相應(yīng)的PCR和基因測序的實驗手冊。
實施例1
1.GLCCI1條件性基因敲除小鼠的制備
1.1打靶載體構(gòu)建
設(shè)計和構(gòu)思基因組中對GLCCI1的打靶原理。GLCCI1打靶載體使用含GLCCI1基因的BAC載體。利用含有GLCCI1的BAC載體套取得到GLCCI1的同源重組臂,通過同源重組的方式將1st-loxp插入exon 2的上游,將2nd-loxp插入exon 2的下游,通過PCR測序及酶切的方式獲得目的打靶載體。構(gòu)建好的載體用PCR及ApaLⅠ、BglⅡ、KpnⅠ、PstⅠ酶切鑒定,產(chǎn)物電泳后照相鑒定條帶。打靶載體PCR鑒定的引物為GLCCI1-FRT-tF:CTGCTTTGAGGTCTTCATCTGC;GLCCI1-FRT-tR:CTTACAGTGCCTCCTCATTACC。
1.2ES細胞篩選及鑒定
將構(gòu)建好的打靶載體進行線性化,線性化的打靶載體質(zhì)粒采用電穿孔的方法注射入C57BL/6N小鼠胚胎干細胞中,使外源DNA與胚胎干細胞基因組中相應(yīng)部分發(fā)生同源重組,將打靶載體中的DNA序列整合到內(nèi)源基因組中從而得以表達。打靶后通過正負篩選、PCR反應(yīng)及Southern blot鑒定得到陽性克隆。用于5’端篩選的PCR反應(yīng)引物為:GLCCI1-CKO-ES1F1:CAGTTCCTGCATTAGCCACCAC;GLCCI1-CKO-ES1R1:CTAGCTTGGCTGGACGTAAACT;產(chǎn)物長度為6040bp者為陽性克隆。用于3’端篩選的PCR反應(yīng)引物為:GLCCI1-LoxP-tF3:acaggaccttacctcacagg;ES-1R4:CTAGAACTAGTGGATCCCCTCGA;產(chǎn)物長度為393bp者為陽性克隆。5’端篩選的PCR反應(yīng)條件為95℃10秒、68℃6分鐘,共40個循環(huán);3’端篩選的PCR反應(yīng)條件為95℃30秒、58℃30秒,72℃40秒,共35個循環(huán)。
用于Southern blot的限制性內(nèi)切酶分別為:5’端為SacI,目標(biāo)片段長度為7.6kb;3’端為PstI+SphI,目標(biāo)片段長度為11.6kb;neo端為ApaI,目標(biāo)片段長度為14.4kb。1.3嵌合小鼠制備
將篩選所獲得的正確同源重組的ES細胞通過囊胚注射獲得嵌合體子代。注射用囊胚采用C57BL/6N近交系小鼠,通過超數(shù)排卵,自然受孕,胚胎體內(nèi)發(fā)育至囊胚階段,用于注射。注射后一如假孕小鼠受體子宮,生育出嵌合率大于50%的嵌合鼠。經(jīng)顯微注射注入假孕鼠子宮中,得到嵌合鼠。嵌合鼠的嵌合率不低于50%。
1.4F1代雜合子小鼠繁育
將獲得的嵌合率高的嵌合鼠與C57BL/6N背景鼠回交,得到F1代雜合子。獲得的F1代雜合子小鼠經(jīng)提取尾基因組DNA進行PCR,用PCR反應(yīng)鑒定基因型。用于F1代雜合子小鼠基因型鑒定的PCR反應(yīng)條件為:95℃30秒;58℃30秒;72℃30秒;35個循環(huán)。用于PCR反應(yīng)的引物分別為:①檢測1st loxP:GLCCI1-FRT-tF:CTGCTTTGAGGTCTTCATCTGC;Neo-5R:ggctggacgtaaactcctc。②檢測2nd loxP:GLCCI1-loxptF3:ctggcagtttgtgaggcatg;ZMK-1R3:AAGGGTTATTGAATATGATCGGA。③檢測內(nèi)參:GLCCI1-loxp-tF:CCCTTGTTTTACATGCGGAGC;GLCCI1-loxp-tR:GCTCACAGTCAGCTAATGGATGG。
1.5獲得GLCCI1-/-純合子小鼠。
將獲得的F1代雜合子小鼠與Flper小鼠配繁一代,去除Neo抗性基因,以排除Neo基因?qū)π∈蟊硇头治龅母蓴_。與Flper小鼠配繁后再和野生型C57BL/6N小鼠回交一代,獲得完全刪去Neo抗性基因的小鼠。再將其與全身性敲除cre品系交配獲得全身性敲除GLCCI1的純合子小鼠。
2.結(jié)果
2.1圖1、圖2為GLCC1條件性基因敲除載體構(gòu)建示意圖。如圖1所示,分別是小鼠GLCCI1基因野生型、打靶突變型基因座位、打靶雙臂位置示意圖。2號外顯子兩側(cè)為同源重組臂,在兩側(cè)的同源重組臂之間,將1st loxP插入2號外顯子上游,將2nd loxP插入2號外顯子的下游,通過PCR測序及酶切的方式獲得目的打靶載體,其中neo為正篩選標(biāo)記,HSV-TK為負篩選標(biāo)記。構(gòu)建好的質(zhì)粒用ApalⅠ、BglⅡ、KpnⅠ、PstⅠ限制性內(nèi)切酶反應(yīng)后電泳鑒定。如圖3所示,T14與DL20為marker,1、2泳道代表ApalⅠ內(nèi)切酶反應(yīng),可切出9836bp,6578bp,1246bp三個條帶;3、4泳道代表BglⅡ內(nèi)切酶反應(yīng),可切出10147bp,5687bp,1200bp,876bp四個條帶;5、6泳道代表KpnⅠ內(nèi)切酶反應(yīng),可切出10182bp,4888bp,2193bp,695bp四個條帶;7、8泳道代表PstⅠ內(nèi)切酶反應(yīng),可切出9984bp,4279bp,2069bp,840bp,525bp五個條帶。圖1-圖3結(jié)果提示檢測片段正確?;虼虬泻螅鯡S克隆鑒定。PCR反應(yīng)鑒定34個ES克隆發(fā)生雙臂同源重組。34個ES克隆PCR電泳圖如圖4A、4B和4C所示。PCR產(chǎn)物經(jīng)Southern blot鑒定進一步證實,最終獲得5個陽性ES克隆。電泳圖如圖5A、5B和5C所示。如圖5A所示,5’端陽性條帶7.6kb見于5A、6B、7C、8D、11G泳道;如圖5B所示,3’端陽性條帶11.6kb亦見于5A、6B、7C、8D、11G泳道。
2.2通過囊胚注射,獲得24只嵌合率>50%的嵌合鼠,出生的嵌合鼠與背景鼠回交后生出的子代經(jīng)PCR基因型鑒定,有4只為雜合子小鼠,為2雌2雄(F1代)。PCR基因型鑒定結(jié)果如圖6所示,其中70、73、77、81為雜合子。
2.3獲得的F1代雜合子小鼠先后與Flper小鼠及野生型C57L/B6N小鼠配繁,獲得完全刪去Neo抗性基因的小鼠。隨后再與全身性敲除的cre品系(CMV-Cre)交配后獲得全身性敲除GLCCI1基因的純合子小鼠。
