本發(fā)明涉及海洋生物醫(yī)藥技術(shù)領(lǐng)域,具體涉及肉芝軟珊瑚的化學(xué)成分提取方法。
背景技術(shù):
肉芝軟珊瑚(sarcophytonsp.),分類學(xué)上屬腔腸動(dòng)物門(cnidaria),珊瑚蟲綱(anthozoa),軟珊瑚目動(dòng)物(alcyonacea),軟珊瑚科(alcyoniidae),肉芝軟珊瑚屬(sarcophyton)動(dòng)物,是一種在熱帶與亞熱帶比較常見的海洋生物。肉芝軟珊瑚顏色多種,包括褐色、綠色或棕色,帶有白色或金色的水螅體,隨著生長會(huì)長出褶皺。珊瑚體呈平鋪的盆形,群體肥厚,中央凹入,表面常具有略呈輻射狀的脊,脊寬約1至3公分;群體的柱部低矮、粗大。
全球有肉芝軟珊瑚約41種,到目前為止,國內(nèi)外研究者已對(duì)該屬的17個(gè)種開展了化學(xué)成分研究。該屬的化學(xué)成分主要有萜類(包括倍半萜、二萜、三萜、四萜,尤其以西松烷二萜居多)、甾醇(尤其以多羥基甾醇居多)、糖苷類、前列腺素、神經(jīng)酰胺、吡啶衍生物及脂肪酸等化合物。這其中不乏許多結(jié)構(gòu)新穎、生理活性顯著的化學(xué)成分,因此我們選擇南海肉芝軟珊瑚(sarcophytonsp.)進(jìn)行化學(xué)成分及其生物活性的研究。
技術(shù)實(shí)現(xiàn)要素:
本發(fā)明旨在提供一種全面提取肉芝軟珊瑚中化學(xué)成分的方法。
肉芝軟珊瑚的化學(xué)成分提取方法,步驟如下:將樣品sarcophytonsp.粉碎后,室溫下用甲醇冷浸提取3次,每次浸泡3天;合并提取液,減壓濃縮,用無水甲醇脫鹽三次,得到總浸膏,總浸膏經(jīng)100-200目的減壓硅膠柱層析,石油醚/丙酮梯度洗脫(100:1到1:2,v/v)。
將上述洗脫的各段產(chǎn)物進(jìn)行進(jìn)一步分離,分離方法包括甲醇/水比例(40%~100%)梯度洗脫,再用半制備hplc進(jìn)一步純化。
本發(fā)明綜合利用tcl、cc、ods、sephadexlh-20、hplc等現(xiàn)代色譜分離技術(shù),從肉芝軟珊瑚(sarcophytonsp.)中分離得到19個(gè)化合物;結(jié)合理化性質(zhì),通過nmr、uv、ir、esims等現(xiàn)代波譜分析技術(shù)等方法技術(shù)鑒定了其結(jié)構(gòu)?;衔锝Y(jié)構(gòu)類型包括13個(gè)多羥基甾醇(sar1-sar13)、3個(gè)西松烷二萜類化合物(sar14-sar16)、3神經(jīng)酰胺類化合物(sar17-sar19)。其中sar1-sar3為新化合物。
具體實(shí)施方式
下面結(jié)合具體實(shí)施例對(duì)本發(fā)明做進(jìn)一步詳細(xì)說明。
樣品來源:
肉芝軟珊瑚(sarcophytonsp.),2010年3月采自中國南海潿洲島海域,水深約12m處,種屬由中國科學(xué)院南海海洋研究所鄒仁林研究員鑒定。標(biāo)本存放于中國海洋大學(xué)醫(yī)藥學(xué)院天然藥物化學(xué)一室,樣品編號(hào)wzd-2010-03。
實(shí)驗(yàn)材料及試劑:
柱層析用ods:北京金歐亞科技發(fā)展有限公司產(chǎn)品;柱層析硅膠(200-300目)、硅膠h以及gf254薄層硅膠、硅膠板:青島海洋化工有限公司;sephadexlh-20:amershampharmaciabiotechab生產(chǎn);
顯色劑:10%h2so4/etoh(v/v);茴香醛-濃硫酸顯色劑;香草醛-濃硫酸顯色劑;
色譜純乙腈和甲醇:美國fisher公司產(chǎn)品;常用有機(jī)試劑:石油醚、環(huán)己烷、氯仿、二氯甲烷、丙酮、乙酸乙酯、鹽酸、甲醇、氯化鈉、氫氧化鈉、三氟乙酸、硝酸銀、濃硫酸、乙酸酐、乙酸、吡啶等均為國產(chǎn)分析純。
樣品的提取分離與純化:
將冷凍貯藏的樣品sarcophytonsp.(濕重5kg)粉碎后,室溫下用甲醇冷浸提取3次,每次浸泡3天;合并提取液,減壓濃縮,用無水甲醇脫鹽三次,得到總浸膏70g。經(jīng)100-200目的減壓硅膠柱層析,石油醚/丙酮梯度洗脫(100:1到1:2,v/v),分為7個(gè)粗組分(fr-1-fr-8)。
對(duì)fr-2,fr-6兩段化學(xué)成分進(jìn)行分離。
組分fr-2(5.4g)的分離
組分fr-2經(jīng)100%~0%石油醚/乙酸乙酯梯度洗脫得12個(gè)組分(fr-2-1~fr-2-12),其中組分fr-2-11再經(jīng)ods柱用甲醇/水比例(40%~100%)梯度洗脫,得5個(gè)組分(fr-2-11-1~fr-2-11-5),組分fr-2-11-2(123.1mg)再用半制備hplc(ymc-packodsc8,10×250mm,5μm;流動(dòng)相:甲醇/水(63:37);流速:2.0ml/min,檢測(cè)波長:230nm)進(jìn)一步純化得化合物sar14(17.3mg),sar15(13.3mg),sar16(4.8mg)。
組分fr-6(4.58g)的分離
組分fr-6粗段活性較強(qiáng)。tlc薄層檢測(cè),10%h2so4/etoh顯藍(lán)色,推斷可能為多羥基甾醇成分,經(jīng)sephadexlh-20柱層析,用甲醇洗脫,去除大分子后,繼續(xù)用硅膠柱層析分離,用石油醚/丙酮(3:1,2:1,1:1)洗脫,得4個(gè)組分(fr-6-1~fr-6-4),其中組分fr-6-2(68.1mg)繼續(xù)ods分離純化,用甲醇/水(30%~85%)梯度洗脫得到3個(gè)組分fr-6-2-1~fr-6-2-3。fr-6-2-3(30mg)進(jìn)一步用半制備hplc純化(ymc-packodsc18,10×250mm,5μm;流動(dòng)相:甲醇/水(90:10);流速:1.5ml/min,檢測(cè)波長:210nm),得單體化合物sar4(2.8mg)。fr-6-3(558.5mg)過反相ods柱,用甲醇/水(30%~85%)梯度洗脫得到4個(gè)組分fr-6-3-1~fr-6-3-4。fr-6-3-2(61.4mg)再通過半制備hplc(ymc-packodsc8,10×250mm,5μm;流動(dòng)相:甲醇/水(80:20),流速:1.5ml/min,檢測(cè)波長:210nm)進(jìn)一步純化得化合物sar5(4.1mg)和sar6(2.2mg)。fr-6-3-3(255.1mg)經(jīng)過sephadexlh-20層析用(petroleumether/ch2cl2/meoh2:2:1)洗脫,得到3個(gè)組分fr-6-3-3-1~fr-6-3-3-3,fr-6-3-3-1(29.1mg)繼續(xù)用半制備hplc分離純化,因?yàn)槎嗔u基甾醇含有多個(gè)羥基的原因,在反相柱上容易拖尾,因此為防拖尾,我們加入1‰的hcooh,流動(dòng)相為83%meoh/h2o:1‰hcooh,(ymc-packodsc8,10×250mm,5μm;流速:1.5ml/min,檢測(cè)波長:210nm),分離純化得到sar7(4.9mg),sar8(3.2mg),sar13(2.2mg),sar17(2.6mg),sar18(1.4mg),sar19(2.2mg)。fr-6-3-3-2(29.1mg)也用流動(dòng)相中加甲酸的方法(ymc-packodsc8,10×250mm,5μm;流動(dòng)相:甲醇/水(80:20):1‰hcooh,流速:1.5ml/min,檢測(cè)波長:210nm)獲得化合物sar9(17.5mg)。fr-6-3-4(105.6mg)直接用半制備hplc(ymc-packodsc8,10×250mm,5μm;流動(dòng)相:甲醇/水(85:15),流速:1.5ml/min,檢測(cè)波長:210nm)分離純化得到單體化合物10(37.3mg)。fr-6-4(1.0g)經(jīng)過正相硅膠硅膠柱層析,因?yàn)榱u基在硅膠柱上太容易吸附,一方面影響分離,另一方面造成樣品損失,所以我們采用三相體系ch2cl2/meoh/h2o進(jìn)行恒度洗脫,比例為300:10:3.3,經(jīng)過硅膠柱層析,分為兩段fr-6-4-1和fr-6-4-2,fr-6-4-1(109.1mg)經(jīng)過半制備hplc(ymc-packodsc8,10×250mm,5μm;流動(dòng)相:甲醇/水(80:20):1‰hcooh,流速:1.5ml/min,檢測(cè)波長:210nm)分離純化得到單體化合物sar1(3mg),sar2(5.1mg),sar11(1.9mg)和sar12(8.7mg)。
