本發(fā)明涉及一些肥厚型心肌病致病基因的新突變,以及它們在肥厚型心肌病治療和診斷方面的用途。
背景技術(shù)
早在19世紀(jì)中葉,法國人A.Vulpian就在鏡下觀察并描述了肥厚型心肌病,但是直到20世紀(jì)50年代后期,肥厚型心肌病獨特的臨床特點才被系統(tǒng)地論述。它通常以不對稱的左和/或右心室肥厚為特征,可累及心室的不同部位,尤以心室間隔為著,左室容積可正?;驕p少。典型的形態(tài)學(xué)改變?yōu)榧±w維排列紊亂。臨床常表現(xiàn)為呼吸困難、心絞痛及暈厥,心律失常、猝死。肥厚型心肌病以往被認(rèn)為是一種少見的心臟疾病,但Maron等進(jìn)行的流行病學(xué)調(diào)查顯示,年輕人的發(fā)病率達(dá)0.2%(Maron BJ,Gardin JM,F(xiàn)lack JM,et al.Prevalence ofhypertrophic cardiomyopathy in a general population of youngadultsechocardiographic analysis of 4111 s ubjects in theCARDIA study.Circulation.1995;92785-789)。我們對中國九省市8080個正常人的超聲心臟多普勒檢查表明,中國人中的肥厚型心肌病亦非常常見,患者可多達(dá)0.08%(Zou Y.等,Am.J.Med.2004,11614-18)。肥厚型心肌病至少有60%-70%以上為常染色體顯性遺傳,稱家族性肥厚型心肌病(FHC)。分子遺傳學(xué)研究發(fā)現(xiàn),編碼肌小節(jié)結(jié)構(gòu)蛋白的基因突變是家族性肥厚型心肌病的根本病因[Seidman JG,Seidman C.The genetic basis for cardiomyopathyfrom mutationidentification to mechanistic paradigms.Cell.2001;104(4)557-67;Bonne,Carrier L,Richard P.Familialhypertrophic cardiomyopathyfrom mutations to functionaldefects.Cir Res.1998;83580-593]。
肌小節(jié)是心肌收縮的基本單位,主要由粗、細(xì)肌絲兩部分組成。粗肌絲主要為肌球蛋白(myosin)和肌球蛋白結(jié)合蛋白C(MyBPC),前者包括兩條肌球蛋白重鏈(MHC)和兩條輕鏈——必需/堿性輕鏈(MELC,MLC-1)及調(diào)節(jié)/磷酸化輕鏈(MRLC,MLC-2)。細(xì)肌絲包括肌動蛋白(actin)、α-原肌球蛋白(α-Tm)和肌鈣蛋白復(fù)合體(TnT,TnC,TnI)。此外,肌小節(jié)成分還有肌連蛋白(titin)等。
1.家族性肥厚型心肌病致病基因1.目前,已證實13個基因160多個位點的突變與FHC的發(fā)病有關(guān),其中有10種是編碼肌小節(jié)結(jié)構(gòu)蛋白的基因,絕大部分突變位于這些基因(Marian AJ,Roberts R.The molecular menetic basis for hypertrophiccardiomyopathy.J Mol Cell Cardio.2001;33655670)(表1)。

然而,這些突變并未囊括所有的遺傳型心肌肥厚型致病基因突變。
本發(fā)明的目的在于發(fā)現(xiàn)新的突變,便于肥厚型心肌病的早期診斷和預(yù)防。

發(fā)明內(nèi)容
本發(fā)明基于的是肥厚型心肌病致病基因中存在一些以前尚未報道的突變這一發(fā)現(xiàn)基礎(chǔ)之上的。本發(fā)明提供了含有這樣的突變的分離核酸。本發(fā)明還提供了可用于檢測這些突變的寡核苷酸,以及利用這樣的寡核苷酸檢測突變的方法。本發(fā)明提供了利用這樣的突變進(jìn)行肥厚型心肌病診斷或者藥物篩選或評估的方法。本發(fā)明另外涉及微陣列。
具體實施例方式
在本發(fā)明中,術(shù)語“樣品”是指來源于人類個體的任何生物學(xué)樣品,包括但不限于例如細(xì)胞,組織樣品如機(jī)體軟組織例如骨組織、毛發(fā)等,生物學(xué)流體如淋巴液、血液、唾液、尿液和其它機(jī)體排泄液等。術(shù)語“分離的”意指非天然存在的?!胺蛛x的核酸”指并不與其所來源的個體基因組中相鄰的5’和/或3’側(cè)翼序列相毗鄰的核酸。因此,“分離的核酸”也包括插入載體內(nèi)的核酸,摻入異源細(xì)胞的基因組內(nèi)的核酸,或者以獨立體存在的核酸。該術(shù)語還包括與其它核酸序列相連接的重組核酸。這樣的重組核酸可能用于生產(chǎn)融合蛋白。
在本發(fā)明中,野生型基因指的是與肥厚型心肌病致病基因?qū)?yīng)的正?;颉K鼈兊男蛄性诂F(xiàn)有技術(shù)中是已知的,并列于例如Genbank數(shù)據(jù)庫中。例如,野生型β-MHC基因的序列以Genbank登錄號giM57965.1公開,在本發(fā)明中以SEQ ID NO1表示;野生型cMYBPC基因的序列以Genbank登錄號giY10629公開,在本發(fā)明中以SEQ IDNO2表示;野生型cTnT基因的序列以Genbank登錄號giX90780公開,在本發(fā)明中用SEQ ID NO3表示。但是,本領(lǐng)域普通技術(shù)人員十分清楚,由于基因的測序過程中可能存在一些錯誤,將會導(dǎo)致野生型基因的序列與本文列出的登錄號指示的序列有所不同,或者核苷酸殘基的編號有所變化。此時,將以最終確定的序列為準(zhǔn)。
本發(fā)明中基因突變的表示方法如下“x nt1>nt2”表示在參考序列中發(fā)生了核苷酸的置換。其中“x”指突變發(fā)生的位點,它遵循的是Genbank中相應(yīng)登錄號給出的野生型基因的核苷酸殘基編號?!埃尽北硎景l(fā)生了置換?!皀t1”指突變前的核苷酸,即正?;?野生型基因)中相應(yīng)位點的核苷酸殘基?!皀t2”指突變后的核苷酸殘基。例如,cTnT基因中的“4693C>T”表示在cTnT基因中發(fā)生了置換突變,導(dǎo)致在相應(yīng)于Genbank登錄號giX90780(SEQ ID NO3)列出的序列中第4693位核苷酸殘基位點處的“C”被取代為“T”。
“x1-x2 del nt1...nt2”表示在參考序列的第“x1”位到第“x2”位的核苷酸位點處發(fā)生了缺失,從而缺失了“nt1”到“nt2”的核苷酸殘基。例如,“14262-14264delAAG”表示第14262位到第14264位的核苷酸“AAG”被缺失了。
“x1^x2 ins nt1...nt2”表示在參考序列的第“x1”位與第“x2”位的核苷酸位點之間插入了“nt1...nt2”核苷酸殘基。例如,“18115^18116insGCAAG”表示在第18115位與第18116位核苷酸位點之間插入了“GCAAG”。
在一個實施方案中,本發(fā)明提供了一種突變的β-MHC基因或其片段,其在對應(yīng)于Genbank登錄號giM57965.1所列序列(SEQ ID NO1)中的下述位置處,含有選自下組的一個或多個突變10353G>A,10429A>C,10895A>T,11811A>C,13653A>C,14449T>C,14452G>A,14557G>A。
在一個實施方案中,本發(fā)明提供了一種突變的cMYBPC基因或其片段,其在對應(yīng)于Genbank登錄號giY10629(SEQ ID NO2)所列序列中的下述位置處,含有選自下組的一個或多個突變4826G>A,8697G>A,8887G>A,12101G>A,13261G>A,14262-14264delAAG,14364delG,18115^18116insGCAAG。
在一個實施方案中,本發(fā)明提供了一種突變的cTnT基因或其片段,其在對應(yīng)于Genbank登錄號giX90780(SE Q ID NO3)所列序列中的下述位置處,含有選自下組的一個或多個突變4693C>T,4769G>A。
在進(jìn)一步的實施方案中,本發(fā)明的突變基因或片段中還含有額外的突變,包括任何已知的或?qū)慝@知的突變或核苷酸多態(tài)性。
本發(fā)明還涉及由上述突變基因或片段衍生的mRNA或cDNA或者編碼的蛋白質(zhì)。
本發(fā)明還涉及可與上述任一突變基因或片段或由其衍生的mRNA或cDNA或者它們的互補序列特異性雜交,但不能與相應(yīng)的野生型基因序列或其互補序列雜交的核酸,例如寡核苷酸,它們可作為探針或引物用于特異性檢測人類個體中的上述突變。本領(lǐng)域中的技術(shù)人員均清楚,探針一般具有與目的核酸的序列互補、并與包含待檢測突變的核酸特異性雜交而不與具有野生型基因序列的核酸相雜交的序列。該寡核苷酸一般定位于突變位點周圍,特異性地檢測樣品中上述突變的存在與否。優(yōu)選地,所述核酸或其互補序列中含有上文所定義的任何一個或多個突變。作為例示,寡核苷酸引物或探針可設(shè)計成待檢測的突變位點位于其5’或3’末端或者在中間,或者待檢測的突變位點位于該引物或探針的互補序列的5’或3’末端或者在中間。在本發(fā)明的上下文中,術(shù)語“特異性檢測”是指該寡核苷酸只與包含上文定義的突變的核酸或其互補序列發(fā)生雜交,而不與分別具有野生型基因序列或其互補序列的核酸發(fā)生雜交。
在突變位點涉及多個核苷酸時,寡核苷酸探針或引物可設(shè)計成使得突變位點的核苷酸序列或與其互補的序列的整個或部分包含在探針或引物的核苷酸序列中。這樣,有可能利用由此制備的寡核苷酸作探針通過雜交或差異雜交的存在與否來測定突變的存在情況,或者作為引物通過擴(kuò)增并隨后檢測(通過例如測序、限制性酶切等)突變的存在情況。
寡核苷酸探針或引物一般長度大于約10bp,例如10-100個核苷酸,優(yōu)選地,其長度為15-75個核苷酸,更優(yōu)選18-50個核苷酸,尤其優(yōu)選為20-35個核苷酸,最優(yōu)選為23-30個核苷酸。本領(lǐng)域中的技術(shù)人員可以很容易設(shè)計出合適的寡核苷酸探針或引物,用于突變位點的檢測。至于其制備,則可通過本領(lǐng)域技術(shù)人員熟知的方法進(jìn)行,包括但不限于例如化學(xué)合成法等。優(yōu)選地,探針/引物是被可檢測標(biāo)記的,以便于檢測。標(biāo)記可以是例如生物素、熒光染料、放射性同位素等,但并不限于此。它們是現(xiàn)有技術(shù)中已知的,并可通過常規(guī)方法連接到探針/引物上。該核酸可以是單鏈的,也可以是雙鏈的。
在本領(lǐng)域中眾所周知,可利用引物通過添加游離核苷酸而延伸核酸序列。雜交指的是在嚴(yán)謹(jǐn)條件下與核酸序列發(fā)生選擇性結(jié)合。在本文中,嚴(yán)謹(jǐn)條件是僅允許引物與含突變的核酸序列或其互補序列雜交,而不與野生型核酸序列或其互補序列雜交的條件。依照所使用的引物,普通技術(shù)人員可以通過常規(guī)實驗確定相應(yīng)的嚴(yán)謹(jǐn)雜交條件。在這方面有眾多的現(xiàn)有技術(shù)可供參考,包括例如Sambrook等,分子克隆實驗室手冊,第2版,冷泉港實驗室出版社,冷泉港,紐約。
本發(fā)明還涉及利用設(shè)計成在擴(kuò)增后將得到包含待檢測突變的片段的引物進(jìn)行突變檢測。所述引物一般定位于突變位點周圍或者其5’或3’方向1000bp、優(yōu)選500bp、更優(yōu)選250bp、尤其優(yōu)選200bp、最優(yōu)選100bp的范圍內(nèi),其長度一般大于約10bp,優(yōu)選為15-100bp,更優(yōu)選為18-75bp,尤其優(yōu)選為20-50bp,最優(yōu)選為25-40bp。
還可以在本發(fā)明的寡核苷酸序列5’和/或3’末端連接一些額外的核苷酸序列。這樣的寡核苷酸也在本發(fā)明的范圍內(nèi)。
利用這樣的寡核苷酸,通過對來自人類個體的遺傳材料進(jìn)行擴(kuò)增和隨后分析,可以檢測所述人類個體中所述突變的存在與否。例如,可以將擴(kuò)增產(chǎn)物進(jìn)行瓊脂糖凝膠電泳,隨后用溴化乙錠、SYBR綠色溶液、諸如此類染色,從而以條帶(DNA片段)的形式檢測擴(kuò)增產(chǎn)物。由此,可以以DNA片段的形式檢測包含突變位點的基因片段。除了瓊脂糖凝膠電泳以外,也可以進(jìn)行聚丙烯酰胺凝膠電泳或毛細(xì)管電泳。進(jìn)一步地,可以分離擴(kuò)增產(chǎn)物,隨后通過測序或者限制性酶切分析突變的存在與否。還有可能使用預(yù)先經(jīng)諸如熒光染料等物質(zhì)標(biāo)記的引物進(jìn)行PCR,并檢測擴(kuò)增產(chǎn)物。還可以采用不需要電泳的檢測方法;在這種方法中,將擴(kuò)增產(chǎn)物結(jié)合到固體支持物諸如微型板上,并通過熒光、酶反應(yīng)、諸如此類等手段來檢測目的DNA片段。
本發(fā)明提供了檢測上述突變的方法,包括應(yīng)用上文所述寡核苷酸探針或引物檢測生物學(xué)樣品,或者應(yīng)用設(shè)計成在擴(kuò)增后將得到包含該突變的片段的引物對擴(kuò)增該生物學(xué)樣品并進(jìn)行檢測的步驟。在檢測中,通常應(yīng)用待測個體的生物學(xué)樣品,例如其血液,但并不限于此。在一個實施方案中,直接應(yīng)用該生物學(xué)樣品進(jìn)行檢測。例如,利用上述核酸作為引物,通過PCR法擴(kuò)增包含待檢測突變位點的片段。在另一個實施方案中,在檢測前首先從該生物學(xué)樣品分離遺傳材料,例如基因組DNA,但并不限于此。檢測可通過例如核酸雜交、PCR法、測序法、限制性片段長度多態(tài)性測定法等方法常規(guī)進(jìn)行,但并不局限于這些方法。其條件是本領(lǐng)域普通技術(shù)人員所知的,或可非常容易地確定。優(yōu)選地,使用如上所述制備的寡核苷酸作為引物,用DNA聚合酶擴(kuò)增編碼包含待檢測突變位點的DNA片段。或者,將如上所述制備的探針與模板DNA雜交,從而檢測那些具有感興趣突變的DNA??梢砸勒粘R?guī)方法來制備模板DNA,如氯化銫梯度離心、SDS裂解法、或酚/氯仿抽提。
本發(fā)明還涉及一種微陣列,其中含有上述寡核苷酸探針或引物。該寡核苷酸可以固定在固相支持物上。在一個實施方案中,所述微陣列是DNA芯片、基因芯片、微型芯片、珠陣等的形式。DNA芯片是將很多種DNA探針排列并固定在基片例如玻璃片上,然后在上面進(jìn)行標(biāo)記DNA的雜交、通過檢測探針上的標(biāo)記信號而可區(qū)分地檢測完全匹配和核苷酸錯配,由此對核酸進(jìn)行檢測。芯片的常規(guī)制備方法是本領(lǐng)域技術(shù)人員已知的。
本發(fā)明還涉及可用于檢測上述突變的試劑盒,其中包括一種或多種相關(guān)的診斷性引物或探針和/或該突變基因特異的寡核苷酸引物,例如前文所述的寡核苷酸探針或引物。試劑盒中還可包括容器、使用說明書、緩沖液、酶和/或其它反應(yīng)成分。優(yōu)選地,其中還包含陽性對照和陰性對照,以便進(jìn)一步確認(rèn)檢測結(jié)果。在一個實施方案中,本發(fā)明的試劑盒包含用于預(yù)定測定法的任何和所有成分酶或成分。這些成分的實例包括但不限于寡核苷酸、聚合酶(如Taq聚合酶)、緩沖液(如Tris緩沖液)、dNTP、對照試劑(如組織樣品、用作陽性和陰性對照的靶寡核苷酸、等)、標(biāo)記和/或檢測試劑(熒光染料諸如VIC、FAM)、固體支持物、說明書、例示性圖表和/或產(chǎn)品信息、抑制劑等。本發(fā)明的試劑盒可以是只包含部分必需成分的部分試劑盒。在這種情況中,使用者可以提供其余成分。本發(fā)明的試劑盒可以包含兩個或多個分開的容器,每個都包含將要使用的部分成分。例如,在試劑盒中,第一個容器包含酶,而第二個容器包含寡核苷酸。酶的具體實例包括適當(dāng)保存緩沖液或容器中的結(jié)構(gòu)特異性切割酶。
本發(fā)明還涉及診斷肥厚型心肌病或確定肥厚型心肌病預(yù)后風(fēng)險的方法,包括對個體的體外樣品檢測下述中的一種或多種突變的步驟β-MHC基因中的下述突變10353G>A,10429A>C,10895A>T,11811A>C,13653A>C,14449T>C,14452G>A,14557G>A;cMYBPC基因中的下述突變4826G>A,8697G>A,8887G>A,12101G>A,13261G>A,14262-14264delAAG,14364delG,18115-18116insGCAAG;或者cTnT基因中的下述突變4693C>T,4769G>A。優(yōu)選地,檢測步驟是按照前文所述突變檢測方法進(jìn)行的。例如,使用位于突變位點上下游的寡核苷酸首先對包含待檢測的突變位點的核酸片段進(jìn)行PCR擴(kuò)增。檢測可通過例如PCR法、測序法、限制性片段長度多態(tài)性等進(jìn)行。
本發(fā)明還涉及利用上述突變基因作為靶點篩選或評估藥物的方法,包括例如對攜帶所述突變的細(xì)胞、組織、器官或動物個體給予一種或多種藥物,并評估其一個或多個指示肥厚型心肌病狀況的指標(biāo),以指示疾病改善狀況。優(yōu)選地,所述細(xì)胞、組織、器官或動物個體是通過基因敲除(gene knockout)或敲入(gene knockin)或者轉(zhuǎn)基因的方法而得到的。基因敲除或敲入或者轉(zhuǎn)基因的方法是本領(lǐng)域技術(shù)人員熟知的,包括例如設(shè)計兩個與內(nèi)源基因同源的側(cè)翼片段。在基因敲除的情況下,兩同源片段之間包含預(yù)定使體內(nèi)內(nèi)源基因失活的突變,從而在導(dǎo)入體內(nèi)后,能將相應(yīng)的內(nèi)源基因失活。在基因敲入的情況下,兩同源片段之間包含預(yù)定導(dǎo)入體內(nèi)內(nèi)源基因處的核酸,從而在導(dǎo)入體內(nèi)后,能將目的核酸插入預(yù)定的位點。操作對象可以是個體,例如小鼠。或者,操作對象也可以是細(xì)胞,例如人的心肌細(xì)胞。這樣,可以在個體或細(xì)胞水平上對藥物進(jìn)行篩選或評估。
下面以實施例的方式對本發(fā)明作出說明。這些實施例都是示范性的,它們不以任何方式限定本發(fā)明的范圍。
實施例實施例1基因組DNA的提取1研究對象的選擇研究對象為在中國醫(yī)學(xué)科學(xué)院阜外心血管病醫(yī)院診斷明確的門診就診或住院的肥厚型心肌病病人,共100例,經(jīng)家族調(diào)查無血緣關(guān)系。其中包括肥厚型心肌病家系先證者51人,相關(guān)家系成員356人,散發(fā)病例49人。正常研究對照120例為性別、年齡匹配的健康人。入選者填寫《分子遺傳學(xué)研究病例調(diào)查表》,內(nèi)容包括病人聯(lián)系方式、個人一般情況、臨床癥狀、體檢結(jié)果、家族史調(diào)查、實驗室檢查、心電圖(必要時Holter),胸片報告,超聲心動圖報告(必要時需心臟MRI或左室及冠脈造影結(jié)果)。建立資料數(shù)據(jù)庫。
肥厚型心肌病診斷標(biāo)準(zhǔn)(WHO/ISFC 1996年公布)(1)基本標(biāo)準(zhǔn)超聲心動圖診斷的原因不明的心臟左室壁厚度大于或等于13mm;或心尖部肥厚或右室肥厚。
(2)參考標(biāo)準(zhǔn)心電圖左室高電壓表現(xiàn),伴/不伴II、III、avF、avL及V4V5導(dǎo)聯(lián)病理性Q波及ST-T改變。
(3)排除標(biāo)準(zhǔn)長期高血壓、缺血性心肌病及其他可引起心室肥厚的原發(fā)性疾病。
(4)家族性肥厚型心肌病確定標(biāo)準(zhǔn)有心肌病的家族遺傳傾向,即病人(先證者)家族中有多人發(fā)病,并符合常染色體顯性遺傳特點。
2基因組DNA的分離取患者肘靜脈血5ml,EDTA(0.5M)抗凝,2500rpm離心10min。小心吸取上層血漿,用1.5ML的Eppendorf管分裝。在血細(xì)胞中加入3倍體積的溶血液(NH4Cl 8.29g、KHCO31.0g、EDTA-Na20.03734g,加雙蒸餾水至1000ml,調(diào)節(jié)PH為7.4),搖勻,冰浴15min。2500rpm離心10min,棄上清。向細(xì)胞沉淀中加入10ml溶血液,搖勻,冰浴15min。3000rpm離心10min,棄上清,得到白細(xì)胞,直接用于提取基因組DNA,或者于-80℃凍存?zhèn)溆谩?br> 如下提取基因組DNA取白細(xì)胞,加4.5ml SE(NaCl 4.383g、EDTA-Na29.306g加雙蒸餾水至1000ml,調(diào)節(jié)PH值為8.0),使勁吹打,混勻。沿管壁加入0.5ml的10%SDS,然后加蛋白酶E(100mg/ml,MilliQ水)50μl,首尾輕搖10min,混勻,然后于37℃水浴過夜。第二天取出,沿管壁加3ml飽和NaCl,迅速手搖15s,使蛋白變性析出,然后于4000rpm離心10min。取上清,轉(zhuǎn)入另一管中,加等體積的三氯甲烷,搖10min,混勻。2500rpm離心10min。取上清,轉(zhuǎn)入另一管中,加等體積的三氯甲烷,搖10min,混勻。2500rpm離心10min,取上清,轉(zhuǎn)入50ml管中,加3倍體積無水乙醇后,首尾輕搖,見DNA絮狀沉淀析出。將DNA挑至管壁,用75%乙醇洗滌2遍。將DNA挑至0.5ml管中,凍干。最后加TE(10mM Tris-HCl、1mM EDTA,調(diào)節(jié)PH為8.0)300μl,使DNA溶解,紫外分光光度計測OD值定量,4℃保存。
實施例2 PCR法篩選β-肌球蛋白重鏈(B-MHC)基因中的突變所用引物及擴(kuò)增條件如下表1所示(基因組序列來自Genebankdatabase,登陸號為M57965)

制備如下PCR反應(yīng)基本體系(50μl)引物(濃度20pmol/μl) 1μl10×buffer 5μlTaqDNA聚合酶(2u/μl) 1μldNTP(10mM) 1μl人基因組DNA 1μlddH2O 41μl按照表1中的PCR條件對各患者或?qū)φ盏幕蚪MDNA進(jìn)行PCR擴(kuò)增,并按下述步驟對PCR反應(yīng)產(chǎn)物進(jìn)行測序前純化步驟(1)將50μl PCR反應(yīng)產(chǎn)物同其6倍體積的無水乙醇與5M醋酸胺(5∶1)混合溶液混合,置于-80℃30分鐘以上。
(2)14000rpm,4℃離心15分鐘,吸上清棄去。
(3)加入200μl的75%酒精,14000rpm,4℃離心5分鐘,吸上清棄去。
(4)室溫或冷凍干燥后,加入20μl TE溶解。
(5)吸2μl點樣,定量。
如下進(jìn)行測序反應(yīng)(1)反應(yīng)體系(20μl)BigDyeTM4μlBigDyeTMBuffer2μl 引物(同PCR擴(kuò)增引物)10pmol/μl(2)反應(yīng)條件(根據(jù)引物的退火溫度而定)

(3)測序產(chǎn)物的純化20μl產(chǎn)物中加25μl醋酸銨和125μl乙醇,置-70℃ 1小時,14,000rpm離心10分鐘,棄上清,加75%的乙醇100μl,14,000rpm離心5分鐘,棄上清,95℃1分鐘烤干,加入2μl loading buffer,混勻,94℃變性5分鐘,然后迅速置于冰上。
(4)將純化后的測序反應(yīng)產(chǎn)物在ABI PRISM377自動測序儀上測序。
(5)用GeneJockey、AssemblyLIGNTM、Sequencing Analysis 3.4軟件閱讀及核對測序圖,并與以公布的基因序列相比較。由此篩選出來的基因突變列于表5中。
實施例3 PCR法篩選心臟肌鈣蛋白T(cTnT)基因中的突變所用引物及擴(kuò)增條件如下表2所示(基因組序列來自Genebankdatabase,登陸號為AY044273)表2

按照表2中的PCR條件對各患者或?qū)φ盏幕蚪MDNA進(jìn)行PCR擴(kuò)增,經(jīng)常規(guī)方法分離擴(kuò)增產(chǎn)物并對其測序。由此篩選出來的基因突變列于表5中。

實施例6限制性片斷長度多肽性(RFLP)檢測因基因某位置的堿基改變,破壞了或建立了可被限制性內(nèi)切酶識別的特異性酶切位點,當(dāng)該樣品被特異性限制性內(nèi)切酶處理后,因其與正常樣品的酶切片段長度不同,在凝膠電泳時,可顯示不同的遷移速度。以此原理,可進(jìn)行已知突變位點的患者家系成員的基因突變篩查。此檢測中使用Promega公司酶切試劑盒,并按照所附的產(chǎn)品說明書進(jìn)行。

2.MHCG425R、Q734P、M822T、G823E;cMYBPCG263R、8887 G>A(intron15)、R668H、G758D、18115^18116 insGCAAG(exon 29);cTnTR145W等突變在家系患者中發(fā)現(xiàn),與肥厚型心肌病呈共分離趨勢。
3.120例正常對照中未發(fā)現(xiàn)以上位點的突變。
