背景技術(shù):
:水稻是重要的糧食作物之一,養(yǎng)活了全球大約二分之一的人口。上個(gè)世紀(jì)60年代,半矮化育種的推廣使得全世界水稻產(chǎn)量得到顯著的提高。近30年以來(lái),雖然水稻雜種優(yōu)勢(shì)利用及其新品種培育在水稻產(chǎn)量提升方面取得了較大的進(jìn)展,但是產(chǎn)量的提高幅度依然不能滿(mǎn)足日益增長(zhǎng)人口對(duì)糧食的需求。預(yù)計(jì)2050年人口數(shù)量將達(dá)到89億,這意味著2050年糧食生產(chǎn)需要提高50%。面對(duì)人口急劇增長(zhǎng),環(huán)境不斷惡化,可耕地面積不斷減少的嚴(yán)峻形勢(shì),如何有效地提高水稻單產(chǎn)已成為農(nóng)業(yè)生產(chǎn)上的一項(xiàng)非常重要的任務(wù)。水稻產(chǎn)量主要由單位面積上的有效穗數(shù)、每穗穗粒數(shù)和千粒重等因素構(gòu)成。分蘗數(shù)決定了有效穗數(shù),分蘗是影響水稻穗數(shù)多少并進(jìn)而影響其產(chǎn)量的重要農(nóng)藝性狀之一;一次枝梗、二次枝梗與結(jié)實(shí)率決定了每穗穗粒數(shù);籽粒大小(長(zhǎng)、寬、厚)決定了千粒重。同時(shí),抽穗期,抗倒伏能力,抗病耐逆特性也影響著水稻產(chǎn)量。水稻穗型是指水稻穗的大小和生長(zhǎng)姿態(tài),是水稻株型的重要組成部分穗粒數(shù)與穗發(fā)育密切相關(guān),小花數(shù)決定了每穗穗粒數(shù);千粒重與谷粒大小和谷粒成熟過(guò)程中灌漿充實(shí)度密切相關(guān),這些農(nóng)藝性狀都與植株產(chǎn)量緊密相關(guān),并且這些重要農(nóng)藝性狀如株高、分蘗數(shù)、分蘗角度、穗部形態(tài)、粒型等都是典型的復(fù)雜農(nóng)藝性狀,受多個(gè)數(shù)量性狀位點(diǎn)(quantitativetraitlocus,qtls)和環(huán)境因子控制。在水稻育種中,理想株型塑造對(duì)水稻品種的遺傳改良發(fā)揮了巨大的作用。傳統(tǒng)的水稻株高較高,莖稈柔弱,對(duì)氮肥耐受力低,易倒伏,產(chǎn)量低;半矮桿基因sd1的引入,使株高顯著降低,莖稈增粗,分蘗數(shù)增多,耐肥與抗倒伏能力提高,因此產(chǎn)量得到顯著提高。為了進(jìn)一步提高水稻單產(chǎn),上個(gè)世紀(jì)80年代國(guó)際水稻所(internationalriceresearchinstitute,irri)提出了理想株型(newplanttype)育種計(jì)劃,即育成少蘗、大穗、莖稈粗壯的理想株型水稻。與此同時(shí),我國(guó)育種家提出了北方粳型超級(jí)稻的理論,指出直立穗育種是繼半矮化育種后適應(yīng)超高產(chǎn)的又一重要的育種形態(tài)進(jìn)化選擇,并育成了以“遼粳5號(hào)”和“沈農(nóng)265”等為代表的一批高產(chǎn)水稻新品種,顯著地提高了我國(guó)粳稻的單產(chǎn)水平。前期,本實(shí)驗(yàn)室從高產(chǎn)粳稻品種“沈農(nóng)265”中克隆了控制該直立穗型性狀的基因dep1,發(fā)現(xiàn)其優(yōu)異等位基因dep1-1能夠增加每穗穗粒數(shù)并提高產(chǎn)量,并已在我國(guó)的“超級(jí)稻”育種中起到了至關(guān)重要的作用。技術(shù)實(shí)現(xiàn)要素:為了進(jìn)一步解析水稻高產(chǎn)株型的分子調(diào)控機(jī)制,我們利用國(guó)際水稻所培育的具有理想株型特征的水稻親本ir66167-27-5-1-6,通過(guò)qtl定位和圖位克隆技術(shù)分離并克隆了控制該理想株型和產(chǎn)量的關(guān)鍵基因npt1。在此基礎(chǔ)上,綜合利用了遺傳學(xué)、分子生物學(xué)、生物化學(xué)等多種手段,深入研究了npt1基因的生物學(xué)功能及其調(diào)控水稻產(chǎn)量的分子基礎(chǔ),為水稻高產(chǎn)育種提供相應(yīng)的理論依據(jù),也為水稻分子設(shè)計(jì)育種提供了重要基因資源。本發(fā)明涉及一種控制水稻莖稈粗度、分蘗數(shù)、每穗穗粒數(shù)、千粒重和產(chǎn)量的理想株型基因npt1及其應(yīng)用。具體地,本發(fā)明涉及npt1在提高農(nóng)作物(例如,水稻、小麥、大麥、玉米、高粱等)的分蘗數(shù)、每穗穗粒數(shù)、千粒重、產(chǎn)量、莖稈粗度和耐倒伏能力方面的新功能和應(yīng)用。更具體地,本發(fā)明涉及的npt1及其同源基因來(lái)源于水稻、小麥、大麥、玉米、高粱等農(nóng)作物。本發(fā)明人利用一個(gè)來(lái)自國(guó)際水稻研究所的少蘗、大穗且莖稈粗壯的水稻親本ir66167-27-5-1-6和我國(guó)粳稻品種春江06雜交所構(gòu)建的重組自交系群體,發(fā)現(xiàn)其中一個(gè)株系ril52表現(xiàn)出了理想株型特征,包括莖稈粗度增加,分蘗數(shù)目減少,單穗粒數(shù)和產(chǎn)量顯著增加等性狀。對(duì)ril52與秈稻品種浙輻802雜交所構(gòu)建的f2群體進(jìn)行了完全隨機(jī)的田間實(shí)驗(yàn),分別考察了分蘗數(shù)、每穗穗粒數(shù)和莖稈粗度這3個(gè)性狀,結(jié)合各性狀的表型值和分子標(biāo)記數(shù)據(jù),qtl分析發(fā)現(xiàn)在8號(hào)染色體長(zhǎng)臂上存在一個(gè)同時(shí)控制莖稈粗度、穗粒數(shù)及分蘗數(shù)的主效qtl,將其命名為qnpt1。隨后利用ril52與浙輻802構(gòu)建的bc2f2和bc2f3群體,將qnpt1精細(xì)定位到水稻第八號(hào)染色體長(zhǎng)臂末端4.1kbp的物理范圍內(nèi),該區(qū)段僅有一個(gè)預(yù)測(cè)的候選基因。在此基礎(chǔ)上,通過(guò)ir66167-27-5-1-6和中花11雜交,并對(duì)目標(biāo)性狀構(gòu)建了中花11背景(zh11)近等基因系,發(fā)現(xiàn)zh11-npt1莖稈更為粗壯,分蘗數(shù)略微減少,株高基本不變,但每穗穗粒數(shù)和千粒重都有明顯增加,單株產(chǎn)量顯著增加。水稻npt1編碼一個(gè)去泛素化酶(osotub1),是人類(lèi)otub1在水稻中的同源基因,因此也將該基因稱(chēng)為osotub1基因,在本發(fā)明中這兩個(gè)術(shù)語(yǔ)縮寫(xiě)可互換使用。研究發(fā)現(xiàn)水稻osotub1(seqidno:15,即水稻ntp1基因編碼的蛋白,在本發(fā)明中也稱(chēng)為npt1蛋白,這兩個(gè)術(shù)語(yǔ)也可以互換使用)具有催化k48位和k63泛素鏈解聚的活性。本發(fā)明人對(duì)osotub1蛋白的亞細(xì)胞定位進(jìn)行了研究,發(fā)現(xiàn)osotub1-gfp融合蛋白定位在細(xì)胞質(zhì)與細(xì)胞核中。另外,構(gòu)建了osotub1自身啟動(dòng)子驅(qū)動(dòng)的posotub1::gus植物表達(dá)載體,通過(guò)農(nóng)桿菌介導(dǎo)的方法獲得了轉(zhuǎn)基因植株。對(duì)轉(zhuǎn)基因植株不同發(fā)育時(shí)期的器官和組織進(jìn)行g(shù)us染色發(fā)現(xiàn),該基因在水稻的整個(gè)發(fā)育時(shí)期都有表達(dá),尤其在維管束系統(tǒng)和分生組織中顯著高表達(dá)。通過(guò)轉(zhuǎn)基因的方法,驗(yàn)證了osotub1基因功能。發(fā)現(xiàn)該基因適度降低表達(dá)或者失去功能突變均會(huì)導(dǎo)致分蘗數(shù)減少,但能增加莖稈粗度、每穗穗粒數(shù)、千粒重,進(jìn)而顯著增加產(chǎn)量。相反,過(guò)量表達(dá)該基因則會(huì)產(chǎn)生類(lèi)病斑表型并引發(fā)細(xì)胞死亡。通過(guò)酵母雙雜交篩選互作蛋白發(fā)現(xiàn)osotub1蛋白能與泛素結(jié)合酶osubc13相互作用,并抑制其功能。進(jìn)一步研究發(fā)現(xiàn)osotub1與水稻株型和產(chǎn)量正調(diào)控因子osspl14蛋白互作,并通過(guò)影響osspl14蛋白穩(wěn)定性來(lái)調(diào)控水稻株型及產(chǎn)量。同時(shí),本發(fā)明人從小麥、大麥、玉米、高粱、小鼠及人類(lèi)中分別克隆到了osotub1同源基因的cdna序列,并通過(guò)轉(zhuǎn)基因研究證明了它們具有與osotub1類(lèi)似的控制水稻莖稈粗度、分蘗數(shù)、穗粒數(shù)、千粒重和產(chǎn)量的生物學(xué)功能。將優(yōu)異等位基因npt1導(dǎo)入高產(chǎn)粳稻品種武運(yùn)粳7號(hào)中,多年多點(diǎn)田間測(cè)產(chǎn)統(tǒng)計(jì)分析發(fā)現(xiàn)npt1和dep1-1聚合能在主栽品種高產(chǎn)基礎(chǔ)上實(shí)現(xiàn)水稻產(chǎn)量進(jìn)一步提升。本發(fā)明的目的在于提供一個(gè)可調(diào)控莖稈粗度、分蘗數(shù)、穗粒數(shù)、千粒重和產(chǎn)量的理想株型基因npt1,為水稻高產(chǎn)分子設(shè)計(jì)育種提供具有重要育種利用價(jià)值的新基因資源。因此,本發(fā)明提供下述:在第一方面,本發(fā)明提供一種水稻理想株型基因npt1及其等位基因npt1,其中npt1能夠增加水稻的莖稈粗度、每穗穗粒數(shù)、千粒重及單株產(chǎn)量。這兩種基因涉及到的核苷酸序列包括:(1)seqidnos:1-4中的任何一個(gè)所示的核苷酸序列;(2)與(1)的核苷酸序列的互補(bǔ)序列在中等嚴(yán)格條件、優(yōu)選高嚴(yán)格雜交條件下雜交的核苷酸序列;(3)與(1)的核苷酸序列具有至少70%、優(yōu)選至少80%、更優(yōu)選至少90%、尤其是至少95%或98%或99%同一性的核苷酸序列;(4)與(1)的核苷酸序列編碼相同氨基酸序列的蛋白質(zhì)、但因遺傳密碼的簡(jiǎn)并性而在序列上不同的核苷酸序列;(5)編碼如下氨基酸序列之一的核苷酸序列:seqidno:15所示的氨基酸序列,或者,由于一或多個(gè)(例如1-25個(gè)、1-20個(gè),1-15個(gè),1-10個(gè),1-5個(gè),1-3個(gè))氨基酸殘基的替代、缺失和/或插入而與seqidno:13所示的氨基酸序列不同的氨基酸序列,或者,與seqidno:15所示的氨基酸序列具有至少70%、優(yōu)選至少80%、更優(yōu)選至少90%、尤其是至少95%或98%同一性的氨基酸序列;(6)(1)-(6)任何一個(gè)的核苷酸序列的活性片段;(7)(1)-(6)任何一個(gè)的核苷酸序列互補(bǔ)的核苷酸序列。在第二方面,本發(fā)明提供第一方面所述的基因及其等位基因的啟動(dòng)子序列,其如seqidno:5或6所示。表1.se 2株系具有莖稈粗度增加,分蘗數(shù)目略減少,穗粒數(shù)顯著增加的理想株型特征的表型(圖1a-e)。將ril52與秈稻品種浙輻802雜交獲得f1,f2后代中隨機(jī)選取196個(gè)單株的葉片提取dna,同時(shí)對(duì)應(yīng)考察倒一節(jié)中部莖稈粗度、分蘗數(shù)和每穗穗粒數(shù)等性狀。利用ssr引物對(duì)ril52和浙輻802進(jìn)行多態(tài)性分析,選取有多態(tài)的引物,分別對(duì)196個(gè)f2單株進(jìn)行pcr擴(kuò)增,根據(jù)電泳結(jié)果和作圖軟件mapmakerversion3.0要求,凡是出現(xiàn)ril52帶型的單株標(biāo)記為b,出現(xiàn)浙輻802帶型的單株標(biāo)記為a,同時(shí)出現(xiàn)ril52和浙輻802帶型的單株標(biāo)記為h。利用winqtlcartographerv2.5及復(fù)合區(qū)間作圖的方法進(jìn)行qtl定位和效應(yīng)估計(jì),結(jié)果發(fā)現(xiàn)在第8號(hào)染色體長(zhǎng)臂上存在一個(gè)同時(shí)控制水稻莖稈粗度(culmdiameter)、分蘗數(shù)(tillernumber)和每穗穗粒數(shù)(grainnumber)的主效qtl,將其命名為qnpt1(圖1f)。將ril52與浙輻802雜交后繼續(xù)回交浙輻802,獲得bc1f2群體;選取類(lèi)似ril52植株表型的單株389個(gè)。進(jìn)一步從中選取表型明確的93個(gè)單株,提取dna進(jìn)行粗定位。利用在目標(biāo)區(qū)域篩選出8個(gè)indel標(biāo)記,將目標(biāo)基因定位于8號(hào)染色體長(zhǎng)臂p6049與p351之間。利用剩余296個(gè)單株進(jìn)行精細(xì)定位,將目標(biāo)基因定位在p5528與p3914之間。將類(lèi)似ril52表型的bc1f2單株與zf802繼續(xù)回交,獲取目標(biāo)性狀的bc2f2分離樣品5326個(gè),繼續(xù)利用在卡位區(qū)間發(fā)展的6個(gè)in-del標(biāo)記篩選交換單株,最終將目標(biāo)基因定位在p139與p143之間的4.1kbp區(qū)間內(nèi),其中包含1個(gè)預(yù)測(cè)基因。該基因編碼一個(gè)去泛素化酶,是人類(lèi)otub1在水稻中的同源基因,將其命名為osotub1(圖2a)。rap-db(http://rapdb.dna.affrc.go.jp/)預(yù)測(cè)osotub1存在兩個(gè)轉(zhuǎn)錄本,其中osotub1.1具有8個(gè)外顯子,7個(gè)內(nèi)含子,編碼區(qū)825bp;osotub1.2具有5個(gè)外顯子,4個(gè)內(nèi)含子,編碼區(qū)597bp。對(duì)卡位區(qū)間進(jìn)行單倍型分析,發(fā)現(xiàn)來(lái)源于ir66167-27-5-1-6的區(qū)段特有4個(gè)snp差異及一個(gè)單堿基插入差異,其中一個(gè)snp位于啟動(dòng)子(圖2b)。qnpt1鑒定與分離所用的分子標(biāo)記是以pcr為基礎(chǔ)的標(biāo)記,包括ssr標(biāo)記和自行設(shè)計(jì)indel標(biāo)記。ssr標(biāo)記均來(lái)自mccouch等(2001,2002)發(fā)表的微衛(wèi)星標(biāo)記連鎖圖;sts標(biāo)記是用ssr分析工具(http://www.gramene.org/gramene/searches/ssrtool)分析克隆序列篩選出微衛(wèi)星重復(fù)性好的ssr目標(biāo)序列,再用primer5分析軟件對(duì)這些目標(biāo)序列進(jìn)行引物設(shè)計(jì)。從這些引物中選擇了75對(duì)在染色體上均勻分布,且在親本間多態(tài)性好的標(biāo)記用于遺傳背景篩選,部分引物名稱(chēng)和序列見(jiàn)表4。pcr程序按照panaud等(1996)的方法稍加修改進(jìn)行,具體為每管20μl擴(kuò)增反應(yīng)體系,包括:0.15μmssr引物、200μmdntps、1×pcr反應(yīng)緩沖液(50mmkcl,10mmtris-hclph8.3,1.5mmmgcl2,0.01%明膠)、50~100ng模板dna,1utaq酶;反應(yīng)程序?yàn)椋?4℃dna變性5min、循環(huán)(94℃1min,56℃1min,72℃1min)36次、72℃再延伸5min。擴(kuò)增出的pcr產(chǎn)物用6%聚丙烯酰胺變性凝膠進(jìn)行電泳,電壓調(diào)至300v,室溫下電泳約3小時(shí)。電泳完畢后,銀染記錄帶型或凝膠成像。以中花11(zh11)的dna為模板,分別擴(kuò)增2.5kbposotub1.1及osotub1.2的啟動(dòng)子區(qū),插入pcambia-2300載體(參見(jiàn)hajdukiewiczetal.,1994),構(gòu)成攜帶水稻自身啟動(dòng)子的載體pcambia-osotub1.1-2300及pcambia-osotub1.2-2300。同時(shí),以zh11的幼穗材料的cdna為模板,擴(kuò)增osotub1.1及osotub1.2的編碼區(qū)序列,分別插入載體pcambia-osotub1.1-2300及pcambia-osotub1.2-2300,構(gòu)成植物表達(dá)載體posotub1.1::osotub1.1及posotub1.2::osotub1.2。通過(guò)農(nóng)桿菌介導(dǎo)的方法轉(zhuǎn)化zh11-npt1,并獲得轉(zhuǎn)基因植株。研究發(fā)現(xiàn)posotub1.1::osotub1.1轉(zhuǎn)基因陽(yáng)性植株一次枝梗和二次枝梗減少,并且穗變長(zhǎng),粒變長(zhǎng),每穗籽粒數(shù)明顯減少,莖稈變細(xì),分蘗增多,類(lèi)似zh11野生型的表型;而posotub1.2::osotub1.2轉(zhuǎn)基因陽(yáng)性植株表型類(lèi)似zh11-npt1。以上結(jié)果表明osotub1.1轉(zhuǎn)錄本具有功能,可以恢復(fù)zh11-npt1的表型。實(shí)施例2:中花11背景近等基因系的構(gòu)建本發(fā)明人將ril52連續(xù)回交中花11,選取莖稈增粗、分蘗減少、穗粒數(shù)增加的植株,并利用indel分子標(biāo)記p135對(duì)目標(biāo)區(qū)段進(jìn)行跟蹤檢測(cè);同時(shí),對(duì)目標(biāo)性狀以外的區(qū)段進(jìn)行背景掃描,最終獲得與中花11遺傳背景十分接近的近等基因系z(mì)h11-npt1材料用于后續(xù)的研究。灌漿后期zh11-npt1表現(xiàn)出莖稈粗壯、有效分蘗數(shù)減少和每穗穗粒數(shù)增加的表型。進(jìn)一步統(tǒng)計(jì)分析發(fā)現(xiàn),相比zh11野生型,zh11-npt1節(jié)間莖稈粗度顯著增加、有效分蘗數(shù)減少大約2個(gè)、穗長(zhǎng)略變短(長(zhǎng)度18-20cm),每穗穗粒數(shù)增加50-70粒,單株產(chǎn)量增加約10%(圖3)。實(shí)施例3:npt1的功能驗(yàn)證分別提取中花11背景的一對(duì)近等基因系幼苗的根(root)、幼嫩莖稈(culm)、葉片(leafblade)、葉鞘(leafsheath)、頂端分生組織(sam)、0.2cm幼穗(youngpanicle)、6cm穗及12cm穗的總rna,反轉(zhuǎn)錄獲取cdna后,定量pcr檢測(cè)osotub1在一對(duì)材料中不同組織和器官中的表達(dá),發(fā)osotub1在水稻整個(gè)發(fā)育時(shí)期都有較高水平的表達(dá),尤其在幼穗中的表達(dá)量高。相比zh11,osotub1在zh11-npt1不同組織和器官中的表達(dá)均呈現(xiàn)出了一定程度的下調(diào)(圖4)。以質(zhì)粒pb1121(chenetal.,2003)為模板擴(kuò)增gus序列,回收gus片段和載體骨架并連接,構(gòu)成載體pcambia::gus-2300;擴(kuò)增2.5kb的osotub1.1啟動(dòng)子序列,酶切回收并插入pcambia::gus-2300,構(gòu)建成posotub1::gus。通過(guò)農(nóng)桿菌介導(dǎo)的方法將其轉(zhuǎn)入zh11,對(duì)轉(zhuǎn)基因植株不同發(fā)育時(shí)期的器官和組織進(jìn)行g(shù)us染色發(fā)現(xiàn),該基因在水稻的整個(gè)發(fā)育時(shí)期都有表達(dá),尤其在維管束系統(tǒng)中顯著表達(dá)。對(duì)生長(zhǎng)一周的幼苗染色,觀察到osotub1在胚芽鞘和根部表達(dá)顯著(圖5a);在根尖分生組織的靜止中心(quiescentcenter,qc)和根冠均有表達(dá)(圖5b);在根部中柱鞘表達(dá)(圖5c);在莖稈維管束有表達(dá)(圖5d);在穗發(fā)育的不同時(shí)期均有表達(dá),到后期表達(dá)逐漸降低,主要集中在穎殼上(圖5e,f)。以pcambia-2300為骨架載體,將osotub1.1啟動(dòng)子插入載體,然后按先后順序插入去掉終止密碼子的osotub1.1編碼序列及gfp編碼序列,構(gòu)建了自身啟動(dòng)子驅(qū)動(dòng)的osotub1.1-gfp融合蛋白植物表達(dá)載體potub1:osotub1.1-gfp并轉(zhuǎn)化zh11。對(duì)轉(zhuǎn)基因幼苗根尖直接壓片觀察,發(fā)現(xiàn)osotub1.1-gfp融合蛋白定位于細(xì)胞質(zhì)與細(xì)胞核中(圖6a)。進(jìn)一步酶解轉(zhuǎn)基因陽(yáng)性植株幼苗葉鞘原生質(zhì)體,也證實(shí)了osotub1.1-gfp融合蛋白定位于細(xì)胞質(zhì)和細(xì)胞核中(圖6b)。以pcambia-2300為骨架載體,構(gòu)建了pactin::osotub1.1過(guò)量表達(dá)載體,并通過(guò)農(nóng)桿菌介導(dǎo)的方法將其轉(zhuǎn)入zh11中,實(shí)時(shí)定量pcr檢測(cè)osotub1.1基因表達(dá)量,獲得了過(guò)量表達(dá)轉(zhuǎn)基因株系oe-8和oe-13。兩個(gè)超高表達(dá)株系灌漿后期表現(xiàn)出穗小、矮化,葉片具有明顯的類(lèi)病斑表型,進(jìn)一步對(duì)出現(xiàn)類(lèi)病斑的葉片進(jìn)行臺(tái)盼藍(lán)染色,發(fā)現(xiàn)部分細(xì)胞出現(xiàn)了細(xì)胞死亡。統(tǒng)計(jì)分析發(fā)現(xiàn)osotub1.1的超高表達(dá)導(dǎo)致一次枝梗、二次枝梗和每穗穗粒數(shù)的顯著減少,株高降低,呈現(xiàn)出劑量效應(yīng)(圖7)。目標(biāo)蛋白的泛素化和去泛素化調(diào)控對(duì)細(xì)胞和生命活動(dòng)發(fā)揮著重要的功能。水稻osotub1編碼一個(gè)去泛素化酶,參與了泛素鏈的解聚。將osotub1.1,osotub1.2構(gòu)建到ge公司原核表達(dá)載體pgex-4t-1上,轉(zhuǎn)入rosetta(de3)菌株中誘導(dǎo)表達(dá)蛋白并純化,購(gòu)買(mǎi)bostonbiochem公司的otub1蛋白及兩種底物泛素鏈tetra-ub。將gst-osotub1.1、gst-osotub1.2和otub1分別與兩種底物泛素鏈tetra-ub于37℃共同孵育1小時(shí),進(jìn)行體外去泛素化實(shí)驗(yàn)。westernblot檢測(cè)發(fā)現(xiàn)tetra-ub-k48與tetra-ub-k63均能夠被osotub1.1裂解,而不能被osotub1.2裂解,這表明具備完整催化活性位點(diǎn)的osotub1.1不僅能夠催化48位泛素鏈的解聚,而且能夠催化63位泛素鏈解聚。進(jìn)一步分析osotub1.1對(duì)k48位和k63泛素鏈的催化活性,發(fā)現(xiàn)二者對(duì)k48位和k63泛素鏈的解聚能力不同,osotub1.1對(duì)k48位泛素鏈的解聚能力更強(qiáng),基本將底物全部解聚,形成ub1;而對(duì)k63位泛素鏈的解聚能力較弱,形成ub3,ub2和ub1(圖8)。實(shí)施例4:npt1/osotub1互作蛋白o(hù)subc13鑒定及功能驗(yàn)證將osotub1.1構(gòu)建在pgbkt7(clontech公司)上,利用酵母雙雜交技術(shù)從水稻穗部cdna文庫(kù)中篩選互作蛋白。采用共轉(zhuǎn)化的方法,初步在-ade/-his/-leu/-trp營(yíng)養(yǎng)缺陷型平板上獲取陽(yáng)性克隆200個(gè),藍(lán)白斑篩選后獲得陽(yáng)性克隆40個(gè)。提取質(zhì)粒重新轉(zhuǎn)化大腸桿菌,證實(shí)了osubc13(os01g48280)、osspl14(os08g39890)分別與osotub1.1相互作用。osubc13編碼一個(gè)泛素結(jié)合酶,其在動(dòng)物中的同源蛋白u(yù)bc13直接與otub1互作,并受到otub1的抑制。將osotub1.1插入pgex-4t-1原核表達(dá)載體(購(gòu)自ge),構(gòu)建成pgex-4t-osotub1.1;osubc13插入pet-28a原核表達(dá)載體(購(gòu)自novagen),構(gòu)建成pet-28a-osubc13。同時(shí)和對(duì)照蛋白gst空載體(購(gòu)自ge)分別轉(zhuǎn)入大腸桿菌菌株rosetta中,誘導(dǎo)表達(dá)并純化獲得gst、gst-osotub1.1與his-osubc13蛋白。將gst、gst-osotub1.1分別與sepharose4b4℃孵育1小時(shí)后,加入his-osubc13蛋白。4℃孵育2小時(shí)后westernblot檢測(cè),gstpull-down實(shí)驗(yàn)證明gst-osotub1.1與his-osubc13相互作用。將osotub1.1連接在psy-735-35s-cyfp-ha(bracha-drori,k.etal,2004)載體,表達(dá)yfpc-osotub1.1;將osubc13連接在psy-736-35s-nyfp-ee(bracha-drori,k.etal,2004)載體,表達(dá)yfpn-osubc13。采用酶法解離7天水稻幼苗第一葉鞘,獲得原生質(zhì)體。通過(guò)peg介導(dǎo)的方法共轉(zhuǎn)化原生質(zhì)體,結(jié)果顯示yfpc-osotub1.1與yfpn-osubc13在細(xì)胞核和細(xì)胞質(zhì)中發(fā)生相互作用(圖9a-c)。以pcambia-2300為骨架載體,分別構(gòu)建p35s:osubc13及p35s::osubc13-rnai植物表達(dá)載體轉(zhuǎn)化zh11,發(fā)現(xiàn)過(guò)量表達(dá)osubc13植株株高無(wú)明顯變化,分蘗略減少,莖稈增粗,一次枝梗和二次枝梗數(shù)目增加,穗粒數(shù)增加,表型類(lèi)似于nil-npt1。對(duì)倒一節(jié)間莖稈中部橫切,發(fā)現(xiàn)莖稈增粗、增厚、維管束數(shù)目增加。而通過(guò)rnai降低osubc13的表達(dá),發(fā)現(xiàn)轉(zhuǎn)基因植株株高無(wú)明顯變化,但分蘗減少、莖稈變細(xì)、一次枝梗和二次枝梗數(shù)量減少、每穗穗粒數(shù)減少,表型類(lèi)似于osotub1.1過(guò)量表達(dá)植株的表型。對(duì)其穗下第一節(jié)間中部橫切,發(fā)現(xiàn)莖稈變細(xì),維管束數(shù)目減少。以上結(jié)果表明osotub1具有抑制osubc13的功能(圖9d-i)。實(shí)施例5:npt1/osotub1互作蛋白o(hù)sspl14鑒定及功能驗(yàn)證osspl14編碼一個(gè)轉(zhuǎn)錄因子,其在rna水平上受到mir156的調(diào)控。osspl14轉(zhuǎn)錄水平和蛋白水平增高,使得水稻分蘗減少、穗粒數(shù)和千粒重增加,同時(shí)莖稈變得粗壯,抗倒伏能力增強(qiáng),進(jìn)而提高產(chǎn)量。為了進(jìn)一步確認(rèn)osotub1與osspl14的相互作用及互作結(jié)構(gòu)域,構(gòu)建了表達(dá)yfpn-osspl14全長(zhǎng)及不同區(qū)段缺失瞬時(shí)表達(dá)載體,分別與表達(dá)yfpc-osotub1.1的表達(dá)載體瞬時(shí)共轉(zhuǎn)化水稻葉鞘原生質(zhì)體。觀察發(fā)現(xiàn)osotub1.1與osspl14的sbp-結(jié)構(gòu)域相互作用(圖10,圖11a)。這表明水稻spl轉(zhuǎn)錄因子的sbp結(jié)構(gòu)域是osotub1.1與osspl14相互作用的關(guān)鍵結(jié)構(gòu)域。免疫共沉淀(co-immunoprecipitation,co-ip)是研究體內(nèi)蛋白相互作用的一種實(shí)驗(yàn)方法。提取zh11背景pactin::otub1.1-gfp轉(zhuǎn)基因植株總蛋白與anti-gfp抗體4℃孵育1h,然后加入40μlproteing瓊脂糖珠后于4℃共同孵育1h。倒掉上清漂洗三次并加入2×sds上樣緩沖液,沸水浴5分鐘后進(jìn)行sds聚丙烯酰胺電泳。westernblot檢測(cè)發(fā)現(xiàn)osotub1與osspl14蛋白在體內(nèi)相互作用(圖11b)。以pcambia-2300為骨架載體,構(gòu)建pactin::osspl14-rnai植物表達(dá)載體轉(zhuǎn)化zh11-npt1,觀察轉(zhuǎn)基因植株發(fā)現(xiàn),降低osspl14表達(dá)水平導(dǎo)致植株分蘗顯著增加,穗部一次枝梗和二次枝梗數(shù)目減少,每穗穗粒數(shù)減少,部分類(lèi)似于zh11表型。將優(yōu)異等位基因osspl14wfp雜交到zh11背景,發(fā)現(xiàn)zh11-osspl14wfp分蘗減少,穗部一次枝梗和二次枝梗數(shù)目增加,單穗粒數(shù)增加,類(lèi)似于zh11-npt1表型。這些研究表明osspl14處于osotub1下游,二者共同調(diào)節(jié)水稻株型(穗型)的形態(tài)建成(圖11c-h)。實(shí)施例6:npt1同源基因的克隆和功能分析在ncbi網(wǎng)站(www.ncbi.nih.nlm.gov)提供的數(shù)據(jù)庫(kù)里,通過(guò)basiclogicalalignmentsearchtool(blast)比對(duì),獲得了小麥、大麥、玉米、高粱、擬南芥、大豆、小鼠及人的同源的cdna序列,分別為烏拉爾圖小麥tuotub1(seqidno:7)、大麥hvotub1(seqidno:8)、玉米zmotub1(seqidno:9)、高粱sbotub1(seqidno:10)、擬南芥atotub1(seqidno:11)、大豆gmotub1(seqidno:12)、小鼠mmotub1(seqidno:13)和人類(lèi)hsotub1(seqidno:14),它們編碼的蛋白的同源性分別為:tuotub1(seqidno:16)同水稻osotub1(seqidno:4)相似性為86.43%;hvotub1(seqidno:17)同水稻osotub1的相似性為86.79%;zmotub1(seqidno:19)同水稻osotub1的相似性為69.38%;sbotub1(seqidno:18)同水稻osotub1相似性為68.10%;atotub1(seqidno:20)同水稻osotub1的相似性為59.80%;gmotub1(seqidno:21)同水稻osotub1的相似性為61.32%;mmotub1(seqidno:22)同水稻osotub1的相似性為38.70%;hsotub1(seqidno:23)同水稻osotub1的相似性為39.12%(圖12)。通過(guò)同源克隆方法分別從玉米、大麥、小鼠和人類(lèi)中分離了osotub1的同源基因zmotub1、hvotub1、mmotub1和hsotub1,分別構(gòu)建了posotub1:tuotub1、posotub1:hvotub1、posotub1:mmotub1和pactin:hsdep1載體,轉(zhuǎn)化zh11-npt1水稻植株;轉(zhuǎn)基因水稻植株表型比較分析顯示過(guò)量表達(dá)玉米、大麥、小鼠及人類(lèi)otub1同源基因均能互補(bǔ)zh11-npt1水稻植株的突變表型,這表明這些同源基因功能具有保守性(圖13)。實(shí)施例7:優(yōu)異等位基因npt1和dep1-1聚合育種應(yīng)用dep1是一個(gè)控制水稻直立穗型和產(chǎn)量性狀的主效qtl,其優(yōu)異等位基因dep1-1能提高水稻莖尖分生組織活性,增加每穗穗粒數(shù),進(jìn)而提高產(chǎn)量(參見(jiàn)本申請(qǐng)人已授權(quán)的發(fā)明專(zhuān)利申請(qǐng),申請(qǐng)?zhí)枮?01110029759.9,在該申請(qǐng)中當(dāng)時(shí)將該基因命名為dep1,后來(lái)為了區(qū)分同一個(gè)dep1基因的不同變異類(lèi)型,將原來(lái)的dep1重新命名為dep1-1)。為了評(píng)價(jià)dep1-1和npt1聚合效應(yīng),通過(guò)雜交的方法將npt1導(dǎo)入攜帶dep1-1(seqidno:33)的高產(chǎn)粳稻品種武運(yùn)粳7號(hào)(wyj7-dep1-1),通過(guò)分子標(biāo)記輔助選擇和連續(xù)回交6次,獲得了wyj7-npt1-dep1-1與wyj7-npt1-dep1-1一對(duì)近等基因系。多年多點(diǎn)田間測(cè)產(chǎn)分析發(fā)現(xiàn),優(yōu)異等位基因npt1和dep1-1聚合不影響水稻
